Studies On Culture Conditions Of Green Vegetative Cell Of Haematococcus Pluvialis

A comparing investigation on the growth situation among three strains of Haematococcus (H. pluvialis CG-11, H. pluvialis CG-16 and H. pluvialis CG-21) cultured in three different mediums (BBM, BG-11 and JM) was conducted, and the cell density, dry weight and the astaxanthin content were measured. The results showed that all the three strains grew better in BBM medium than in BG-11 and JM. Among three strains, H. pluvialis CG-11 cultured in BBM medium grew best, the cell density, dry weight and the astaxanthin content were higher than the other two strains, and the final cell density reached to 59.8×104 ells·ml-1, the dry weight was 0.527g·L-1. The astaxanthin content was 3.55mg·L-1. It was proved that H. pluvialis CG-11 was suitable mass culture. Using H, pluvialis CG-11 as test material, and BBM as culture medium, the effects of different nutrients and environmental conditions on growth, chlorophyll a content, carotenoid content, dry weight and astaxanthin content of the green vegetative cell were investigated. The results showed:(1) Comparing the growth of H. pluvialis CG-11 cultured in different concentration of BBM medium, the best growth was found in the 1 × BBM medium, the cell density of H. pluvialis CG-11 reached the highest value of 49.3×104 cells·ml-1 at the 18th day of.culturing(2) Under the same N concentration, the growth situation of H. pluvialis CG-11 in different N sources (NH4Cl, NaNO3, NH4NO3 and NH2CONH2) in BBM medium were compared, it was found that the NH4Cl was not good for its growth. The nitrogen source of both NaNO3 and NH4NO3 were found to be suitable to cell growth. The dry weight, chlorophyll a content, carotenoid content and astaxanthin accumulation were higher in these two N sources than in other N sources.(3) Using NaNO3 and NH2CONH2 as the nitrogen source, the effects of different N concentration on the growth of the H. pluvialis CG-11 were studied. The results showed that the suitable N concentration for the growth of the algae was 41.2 mg·L-1, and the dry weight, chlorophyll a content, carotenoid content and the astaxanthin content were significantly higher than in other N concentration levels, any level higher or lower than 41.2 mg·L-1 was less better for H. pluvialis CG-11 growth.(4) The study of different phosphor concentration effects on growth showed that the...
