Biothiol is a most special type of amino acids which contain one sulfhydryl groups in the molecular structure and plays an important role in human's metabolism and information transmission in vivo.Cysteine(Cys)is the most abundant amino acid in biothiol,as one of the essential amino acids in our body,which is involved in the synthesis of polypeptides and proteins in organisms.Recent studies have shown that Cys plays an important role in pathology and physiology: studies have shown that changes in the content of Cys in organisms are closely related to the occurrence and development of some important diseases,because of abnormal changes in Cys content have been detected in organisms suffering from Alzheimer's disease,Parkinson's disease and other diseases.The detection of Cys in vivo is essential for studying its roles.Fluorescence-based detection methods have attracted much attention for the significant advantages these offer to the life sciences,including nondestructive detection,excellent selectivity,and high sensitivity.Herein,we designed and synthesized a novel coumarin-based fluorescent probe 1 and a novel phenothiazine-based fluorescent probe 2.The probe 1 could be applied to monitoring Cys in living He La cells,which indicates that this probe could penetrate viable cell membranes and image Cys.Phenothiazine,the main structure of probe 2,has been used as a Cys probe in some cases,but in this study the probe 2 don't show a recognition effect on biothiol.In this study,a fluorescent probe was successfully constructed for the in vitro imaging of Cys.The probe could detect Cys in solution and image endogenous Cys in living He La cells.This coumarin-based probe exhibits ideal characteristics including rapid detection,good selectivity,and good membrane permeability.The linear relationship of Cys in the range of 0-50 ?M,provides a potential possibility for the quantitative study of intracellular Cys.By detecting changes in the content of Cys in the body,it can provide a correct basis for the diagnosis of certain diseases in the body.Therefore,it is critical to detect and distinguish Cys in vivo,which has great significance for studying its function in physiological activities.
