In this paper,a series of organic small molecule fluorescent probes based on coumarin-rhodamine were designed and synthesized,and their properties were studied.The main contents were included as follows:1.The development history,luminescence mechanism and relevant parameters of fluorescence are introduced.The composition of common fluorescent probes is described and the main recognition mechanisms of fluorescent probes are summarized.2.Rhodamine B thionyl hydrazine and coumarin derivatives were chosen as precursors,and two Hg2+fluorescent probes CRS1 and CRS2 based on through bound energy transfer(TBET)system were designed and synthesized,and their structures were characterized.The recognition mechanism,optical properties and theoretical calculation model of two probes for the detection of Hg2+were systematically studied.Furthermore,CSR1 and CSR2 were successfully applied to image Hg2+in living He La cells and in vivo.3.Three Cl O-fluorescent probes CRO,CRO1 and CRO2 were designed and synthesized using rhodamine B hydrazide and coumarin derivatives,and their structures were characterized.The dihydrazide structure in the probe can specifically recognize Cl O-,and is oxidized by hypochlorous acid to rhodamine fluorophore and coumarin derivative fluorophore.In this paper,the optical properties of CRO,CRO1 and CRO2 were tested.At the same time,the detection of exogenous and endogenous Cl O-by probe CRO was successfully applied to flow cytometry experiments of He La cells and Raw264.7 cells respectively.The recognition of exogenous and endogenous Cl O-by probe CRO1 was successfully applied to fluorescence imaging by laser scanning confocal microscope of He La cells and Raw264.7 cells respectively.The identification of exogenous Cl O-by probes CRO1 and CRO2 was successfully applied to the in vivo imaging experiment of mice.In summary,we designed and synthesized a series of coumarin-rhodamine fluorescent probes.The structure of the probe molecules was characterized,their optical recognition and detection performance of the probes on target molecules were analyzed,and the reaction mechanism of the recognition were studied.The biological application performance of the probes on target molecules were further expanded.
