With the deepening of modern life science research and the rapid development of new interdisciplinary research fields,high-resolution microscopy has become an indispensable scientific research technology in life science research.High resolution microscopy can be divided into two categories:wide field fluorescence microscope and optical slice fluorescence microscopy.it can be used to obtain clear high quality and high resolution fluorescence images,and can be used to observe fixed cells,living cells and so on.High resolution microscope can obtain clear and sharp multi-layer Z plane structure,time series image,large field pictures,etc.,and can carry out fluorescence co-localization,FRAP and other quantitative analysis,as well as molecular number and brightness analysis.In this paper,many types of high resolution optical microscopy,including general laser confocal,structure optical,Apotome,Airyscan technique,and these methods are used to compare the imaging of animal and plant biological samples.In this paper,different types of objective lenses with different magnification are studied to image the selected samples,and the effects of fluorescent images are analyzed and explored.The objective lenses studied include APO objective lenses,LD lenses,large field objective lenses and so on.The optical microscopic imaging equipment includes upright microscope,inverted fluorescence microscope,stereo microscope and so on.In this paper,the mainstream high resolution optical slice fluorescence method is compared with the wide field fluorescence imaging method,and the advantages and limitations of different methods are analyzed by practical examples.At the same time,this paper involves the research of some super resolution optical microscope imaging,and the super resolution optical microscope is studied and analyzed through the imaging case.
