PurposeFiltering out METTL8--the potential biomarkers and immunotherapy targets for lung squamous cell carcinoma based on Bioinformatics,and verifying the correlation between METTL8 and CD8~+T cell immune infiltration.Method1.The research is based on big data of bioinformatics.TCGA database extracts the original m RNA expression processing data of LUSC.GEO database downloads the GSE17710 data set.WGCNA software package constructs weighted gene coexpression network analysis.GO database and KEGG database are used for enrichment analysis.TIMER database assesses the level of immune cells in tumor tissues.META analysis and KM-plot database are used for survival analysis.In addition,there are PPI,GSEA enrichment analysis,and Cbioportal database to assist in screening genes.2.Detecting the differential expression of METTL8 and CD8~+T cells in LUSC tissue samples by IHC,and to verify the correlation between METTL8 and immune infiltration of CD8~+T cell.3.Combining the expression levels of METTL8 and CD8~+T cells in different LUSC samples to analyze their correlation with clinical manifestations.Results1.Analyzing the dataset GSE17710 through GEO database,and extract the brown module from it.Founding that this set of differentially expressed genes is enriched in a variety of migration and invasion related functions and cancer signaling pathways through GO and KEGG enrichment analysis.Then,screening out 8 HUB genes through protein-protein interactions(PPI)network.Finally,use a variety of databases to screen for different requirements,and get the required gene METTL8.2.The expression level of METTL8 in LUSC samples was significantly higher than that in normal tissues and adjacent tissues.The expression level of CD8~+T cells in samples was negatively correlated with METTL8.3.Statistical analysis found that the expression of METTL8 and CD8~+T cells are different in LUSC tissues of smoking history.Conclusion1.METTL8 has inhibitory effect on CD8~+T cell immune infiltration.2.METTL8 may be identified as a potential biomarker and immunotherapy target in LUSC.
