Background and purposeOvarian cancer is one of the three major gynecologic malignancies,and the mortality rate is the first among gynecologic malignancies.The symptoms of early ovarian cancer were not obvious,and the diagnosis was mostly late,while the 5-year survival rate of the patients with advanced ovarian cancer was only 29%.Therefore,early detection and early diagnosis of ovarian cancer are very important to improve the survival and prognosis of patients.ENTPD5 is a nucleotide hydrolase encoding the endoplasmic network,which can be regulated by mutation p53 and participate in the occurrence,development and metastasis of tumor.ENTPD5 has been proved to be highly expressed in cervical?prostate?breast?brain and other diseases.However,there is no report on the expression of ENTPD5 in ovarian cancer.Based on the bioinformatics method,this study collects several database information mainly based on Oncomine,explores the expression of ENTPD5 in epithelial ovarian cancer and expounds the possible mechanism of its participation.Then,the expression of ENTPD5 in epithelial ovarian cancer is verified by the temporary specimen.This will not only reveal the legacy mediated by ENTPD5.The information transmission network provides important molecular basis for the early diagnosis and early treatment of ovarian cancer,and also provides a new potential target for tumor treatment.Methods1.Bioinformatics analysis(1)The expression level of ENTPD5 in epithelial ovarian cancer was analyzed by Oncomine and TCGA database;(2)The relationship between the expression level of ENTPD5 and clinical characteristics of patients with epithelial ovarian cancer was analyzed by UALCAN database;(3)Gene enrichment analysis(GSEA)software was used to analyze the pathway of ENTPD5 in epithelial ovarian cancer;(4)The correlation between ENTPD5 and immune cell content in epithelial ovarian cancer was analyzed by Cibersort package.2.Basic experimental verification(1)The expression of ENTPD5 in 23 epithelial ovarian cancer tissues and 15 normal ovarian tissues was detected by RT-qPCR;(2)The expression of ENTPD5 in 23 epithelial ovarian cancer tissues and 15 normal ovarian tissues was detected by Western Blot;(3)Immunohistochemistry was used to detect the expression of ENTPD5 in 50 paraffin embedded epithelial ovarian cancer samples and 6 paraffin embedded normal ovarian samples.Result1.Bioinformatics analysis(1)The expression of ENTPD5 in epithelial ovarian cancer was analyzed by Oncomine and TCGA database.The results showed that the expression of ENTPD5 in epithelial ovarian cancer was significantly higher than that in normal ovarian tissue(P<0.05);(2)The expression level of ENTPD5 was found to be related to the age of patients;(3)Through GSEA software analysis,it was found that ENTPD5 was involved in ABC transporter,WNT signaling pathway,insulin signaling pathway and glycolysis,and highly enriched in them;(4)Cibersort analysis showed that the expression level of ENTPD5 was negatively correlated with the content of NK cells,mast cells and eosinophils(P<0.05).2.Basic experimental verification(1)The expression of ENTPD5 in normal ovary and epithelial ovarian cancer was compared by RT-qPCR.The results showed that the expression level of ENTPD5 in epithelial ovarian cancer(2.592±0.339)was significantly higher than that in normal ovary(1.00±0.123)(t=4.312,P<0.01);(2)The expression of ENTPD5 in normal ovary and epithelial ovarian cancer was compared by Western Blot.The results showed that the expression of ENTPD5 in epithelial ovarian cancer was significantly higher than that in normal ovary(P<0.05);(3)High expression of ENTPD5 was found in epithelial ovarian cancer by immunohistochemical staining(P<0.05).ConclusionThrough bioinformatics analysis was found that ENTPD5 was highly expressed in epithelial ovarian cancer tissues,which may promote the occurrence and development of ovarian cancer by participating in B and T cell-mediated signaling pathway?ABC transporter mediated signaling pathway?WNT signaling pathway?insulin signaling pathway?glycolysis process?tumor malignant transformation process and tumor cell immune infiltration.Based on the research results of bioinformatics,further adoption RT-qPCR?Western Blot and immunohistochemistry results showed that ENTPD5 was highly expressed in ovarian cancer and provides a basis for further study of ENTPD5 in ovarian cancer,and reveals that ENTPD5 may become a new target for early diagnosis and treatment of ovarian cancer.
