| ObjectiveNID2 is involved in the occurrence and development of a variety of tumors,but its role in gastric cancer is not yet clear.This study used bioinformatics and immunohistochemistry methods to study the expression of NID2 in gastric cancer(GC)and its clinical significance.In order to explore whether NID2 has the potential to become a new target for the treatment of gastric cancer.MethodsFirst,analyze the difference between NID2 in gastric cancer and normal tissues through Oncomine online database.Then use the Linked Omics online database to find its positively related genes and import them into the David database for functional annotation and pathway analysis.Furthermore,the Protein Interaction Network(PPI)was built through the String database and Cytoscape software.The GEPIA database was used to analyze the relationship between the expression of NID2 and different stages of gastric cancer and the relationship between the survival and prognosis of patients.The TIMER database was used to study the relationship between NID2 and immune cell infiltration.Use the UALCAN database to explore the methylation of NID2 in gastric cancer.Finally,immunohistochemical method was used to verify the expression of NID2 in gastric cancer.ResultsThe expression of NID2 in gastric cancer was significantly higher than that in normal tissues(P < 0.05).Functional enrichment annotation and pathway analysis show that NID2 and its positively related genes are mainly related to collagen fiber tissue,cell response to amino acid stimulation,cell adhesion,ECM-receptor interaction,protein digestion and absorption,focal adhesion,PI3K-Akt signaling Pathways,amoebiasis,etc.The GEPIA database showed that the expression level of NID2 in gastric cancer was significantly increased(P<0.05),and there were significant differences in different stages(P<0.05).The survival rate of the high expression group was significantly lower than that of the low expression group(P<0.05).NID2 may also be related to the infiltration of immune cells.The results of immunohistochemistry showed that the positive rate of NID2 in gastric cancer tissues was 92.6%,which was significantly higher than that of adjacent tissues of 51.8%,but it had nothing to do with gender,age,tumor size,lymph node metastasis,stage and degree of differentiation.ConclusionsNID2 may be involved in the occurrence and development of gastric cancer,and immune cell infiltration is related to the prognosis of patients.It is expected to become a molecular target for early diagnosis,treatment and prognosis of patients with gastric cancer. |
