PCR-polymerase chain reaction is an in vitro nucleic acid amplification technology,which can achieve the purpose of rapid detection and help people discover unknown diseases in advance.It is mainly used in medical diagnosis,food safety testing,forensic identification,gene cloning and other molecular sciences.Basic research and application analysis.The main design of this subject is a 96-well slot-type nucleic acid amplification instrument,which can achieve the purpose of less samples,rapid amplification,and rapid detection.The main research contents of this article are as follows :(1)Selection and design of the key components of the variable temperature system.First,according to the requirements of the PCR thermal cycle system,a comparison of different resistance temperature sensors was carried out,and the resistance temperature measurement and resistance temperature measurement circuit suitable for the system requirements were selected.Secondly,the calculation on TEC selection was carried out,and the appropriate TEC and quantity were selected.Finally,the heat dissipation structure required in the system was designed.Through the simulation analysis of the different heat dissipation rack structures designed,the heat dissipation rack suitable for installation and system requirements was selected,so that the whole device was kept simple and the response aspect was achieved.Requirements.(2)Analysis and research on the uniformity and delay of the reaction device.During the reaction of the system,the flow of the sample block and the surrounding air will cause the loss of heat during the heat transfer process,resulting in uneven heat transfer.This subject was simulated and analyzed by the finite element software ANSYS,and proposed Measures are taken to improve the unevenness in the heat transfer process,avoid the adverse effects of heat loss,and at the same time simulate and analyze the delay in the heat transfer process to achieve the purpose of precise control.(3)Design and experiment of temperature control algorithm.For different control schemes,the subject chooses a control scheme that combines PID control and fuzzy control,and conducts experimental comparisons.It is concluded that under the control algorithm that combines PID control and fuzzy control,it has small overshoot and fast speed.Response speed,thermal cycle control system can meet the requirements of various control indicators..Through the selection and design of the subject PCR instrument and research and study,the amplification system obtained can meet the relevant requirements and achieve a good reaction purpose.However,there are specific aspects such as detection and temperature error accuracy that need to be improved in design.
