Fatty acids are essential for insect growth and development and maintaining normal physiological activities of insects.Fatty acid synthase system(FAS)is an enzyme system that plays a key role in the process of insect fatty acid synthesis.Among them,acyl carrier protein(ACP)and malonyl/acetyl transferase(MAT)are important components of FAS.In order to further clarify the functions of ACP and MAT genes,this study used the environmental resource insect Hermetia illucens as experimental material and through gene cloning,q RT-PCR technology,RNAi technology,etc.,to study the expression characteristics and functions of H.illucens HiACP and HiMAT genes.In order to explore the molecular mechanism of the above-mentioned genes regulating the fatty acid synthesis of H.illucens,clarify the influence of silencing target genes on the growth and development of H.illucens,and lay a theoretical foundation for the use of H.illucens to produce high-quality fatty acids.The main experimental results obtained in this paper are as follows:The full length of open reading frame of HiACP and HiMAT genes of H.illucens was obtained by amplification.The results of spatiotemporal expression showed that the relative transfer rate of HiACP and HiMAT genes was the highest in egg stage and larvae intestine,which was significantly different from other stages of development and tissue(P<0.05).The results showed that the expression of HiACP and HiMAT genes was specific in different developmental stages and different parts.RNAi technology was used to silence HiACP and HiMAT genes of H.illucens.The results showed that the best silencing efficiency was obtained by injecting 2000 ?g/m L ds RNA on the third day.HiACP and HiMAT gene silencing significantly increased the mortality of the 3rd instar larvae(P<0.01).After HiACP gene silencing,the proportion of saturated fatty acid(SFA)in the larvae decreased,the percentage of C12 decreased by 3.26%(P<0.01),and the proportion of unsaturated fatty acid(UFA)increased.After HiMAT gene silencing,the proportion of SFA decreased,the percentage of C12 decreased by 5.66%(P<0.01),the proportion of UFA increased,and the fatty acid composition changes,in which C15:0 and C20:0 missed.These results suggest that HiACP and HiMAT genes can regulate fatty acid synthesis in the larvae of H.illucens.HiACP and HiMAT genes of H.illucens were silenced to regulate the synthesis of fatty acids,which inhibited the growth and development of H.illucens.The results showed that the growth of the 3rd instar was slow and the body weight and length of the larvae were decreased after HiACP and HiMAT gene silencing.The results showed that the larvae instar increased,the adult body weight,body length,adult eclosion rate and oviposition of each female adult decreased significantly(P<0.01).These results indicate that HiACP and HiMAT genes play an important role in the growth and development of H.illucens.
