Objective To analyze the gene expression profile chip of anaplastic thyroid carcinoma(ATC)by bioinformatics method,to mine the key signaling pathways and Hub genes in the occurrence and development of ATC,and look for specific molecular markers and potential therapeutic targets.Methods Download the gene expression profiles chip of ATC from the GEO database of National Biotechnology Information Centre,the limma package in the R language was used to filer the differential expression genes(DEGs)between ATC and normol thyroid tissue.The GO enrichment analysis and KEGG pathway analysis of the DEGs were performed by using DAVID database.Construct and visualize the protein interaction network in the STRING database and Cytoscape.Results 417 differentially expressed genes were obtained,of which 124 were up-regulated and 293 were down-regulated.Through Go enrichment,we found that: the DEGs were significantly involved in the biological process of oxidation-reduction process,cell division,mitotic nuclear division,cell proliferation and chemotaxis.Analysis of KEGG signaling pathway shows that the DEGs are mainly enriched in the Interaction of cytokine receptors,cancer signaling pathways,systemic lupus erythematosus,PI3K-Akt and HIPPO signaling pathways.The 10 hub genes such as MYC、CDK1、CCNA2、AURKA、CCNB1、FN1、AURKB、PLK1、KIF11、CDC20 were screened by constructing a protein interaction network.Conclusion This study obtains 10 key genes related to the occurrence and development of ATC,these genes can potentially provide novel strategy for the treatment of ATC.
