Objective:(1)Summarize the different plants of Paris genus in Yunnan Province,and supplement the botanical characters.(2)We use chemical morphological analysis combined with DNA barcode technology to sort out the varieties and distribution of Paris plants in Yunnan Province.(3)This paper discusses the genetic variation and genetic relationship of different types of Pais polyphylla Smith var.yunnanensis(Franch.)Hand.-Mazz.(4)Objectively evaluates the quality of Paris genus medicinal materials.It is expected to provide a scientific basis for the protection,rational development and utilization of Paris genus resources.Methods:(1)Through the literature review and field investigation,the morphological data of plant samples were collected to establish the morphological data of the Paris genus.(2)Based on the morphological taxonomy,systematically sort and revise the species definition of Paris medicinal plants through DNA barcodes(ITS sequences).(3)Establish a DNA barcode database of two traits of Paris polyphylla var.yunnanensis,and then explore the genetic relationship at the molecular level,which provides a powerful complement to whether different varieties of Paris polyphylla var.yunnanensis can be classified into one category.(4)Simultaneous determination of six saponins(paris saponin I,II,III,VI,VII and saponin)in19 species of Paris by HPLC.Results:(1)According to the “Li Heng's” classification system,combined with DNA barcode technology,there are currently 19 species and 15 varieties of Paris genus in Yunnan Province.New species of Paris caojianensis and Paris gaoganensis were found,and a suspected new species(Paris sp.nov.)(2)Supplement and revise the morphological traits and species distribution of 10 species,7 varieties,3 new species,with the color maps of each species were added.(3)Combining morphology and DNA barcode technology,it is proved for the first time that Paris kwantungensis should be classified as a subspecies of Paris polyphylla var.chinensis.In addition,there is a new subspecies Paris gaoganensis under Paris polyphylla var.yunnanensis,and the traditional Paris polyphylla var.yunnanensis with shorter stems.(4)DNA barcodes can effectively identify 24 species of Paris genus plants,and this study supply the molecular data of Paris caojianensis,Paris tengchongensis and other species.(5)The saponin content of Paris forrestii,Paris axialis,Paris tengchongensis etc.meet the standard of China Pharmacopoeia,and can be considered as alternative medicinal materials.Conclusion:In this paper,we established the plant resources database of Paris genus in Yunnan Province for the first time,and discovered two new species and one new variety.Combining morphology and DNA barcode technology to clarify the species of Paris kwantungensis.In addition,we systematically studied the morphology,DNA barcode and quality evaluation for 19 species of the Paris genus,and it is recommended that Paris forrestii,Paris axialis and Paris tengchongensis could be the potential alternative sources of medicinal materials.The results of this study not only provides data basis for the diversity of Paris plants in Yunnan Province,but will lay the scientific foundation for the comprehensive protection,rational development and utilization of Paris resources.
