Morphometrics and DNA barcodes of some species of Lasiini and Plagiolepidini were studied.Lasiini and Plagiolepidini were classify and identify using two methods of morphometrics analysis and five analysis methods of DNA barcodes.The molecular phylogenetic relationship of these species were built by using Maximum likelihood(ML)and Bayesian Inferences(BI)methods.Tapinoma melanocephalum was used as outgroup.The main results were as follows:1.The barcode sequences of COI were analyzed using Maximum Likelihood(ML)and Bayesian Inferences(BI)mothods on 44 species of 9 genera of Lasiini and Plagiolepidini.The results were as follows: In the category of genera,Lasius and Prenolepis are monophyletic groups.In the category of species,the results support the inclusion of Paratrechina umbra in Nylanderia;Lepisiota rothneyi in Plagiolepis.2.Classification and identification of Lasiini and Plagiolepidini ants based on morphometrics and DNA barcodes(1)Morphometry dataset was classified and identified using artificial neural networks and Gaussian mixture model analysis methods for 28 species of ants(including outgroups).The results showed that the identification success rate of artificial neural network analysis was 86.35%,and the absolute identification success rate was 36.86%;the identification success rate of Gaussian mixture model analysis was 91.46%,and the absolute identification success rate was 65.19%.(2)The COI gene barcode sequence was classified and identified using NJ,fuzzy Id,BCM,j MOUT and ABGD analysis methods for 45 species of ants(including outgroups).The results showed that the identification success rates of NJ,fuzzy Id and BCM analysis methods were 100%,99.59% and 100%,respectively;the relative identification success rate was 98.84% and absolute identification success rate was 86.54% of j MOUT and ABGD methods.All have strong ability of species identification.(3)The comparison classification and identification of 27 species based on DNA barcoding and morphometrics.The results showed that: 26 species of ants were successfully identified,including Nylanderia flaviabdominis,Pseudolasius silvestrii,Lasius niger,Paraparatrechina sauteri,Prenolepis naoroji,Prenolepis cyclopia,Lepisiota rothneyi,Nylanderia amia,Lasius nipponensis,Lasius flavus,Paraparatrechina butteli,Prenolepis angularis,Plagiolepis manczshurica,Paratrechina umbra,Nylanderia bourbonica,Nylanderia flavipes,Nylanderia pieli,Lasius fuliginosus,Lasius talpa,Paraparatrechina sp2,Nylanderia sp2,Pseudolasius risii,Lasius alienus,Nylanderia integer,Pseudolasius emeryi,Tapinoma melanocephalum,and the identification results were consistent with the morphological classification.The results show that the DNA barcode technologies have high identification success rate in Lasiini and Plagiolepidini,and is more accurate than the results of morphometric studies.
