Purpose: To investigate the association between FTO polymorphism and aerobic capacity evaluation index,initial value of body composition and blood metabolism index and exercise fitness effect of Han Chinese people in order to screen out the molecular genetic markers for the difference of predicting individual and exercise fitness effect,and apply them to the formulation of public fitness plan.Methods: Based on the study of the science and technology supporting subject's sample and DNA database,476 healthy people,age 20-69,who had not participated in regular physical exercise for one year were selected.The main testing index including: VO2 max,BMI,percentage of fat,triglyceride,total cholesterol,low-density lipoprotein,high-density lipoprotein and etc.And they were used to express the exercise fitness effect after exercise intervention.The exercise mode was:climbing,running,power bicycle and comprehensive strength exercise intervention.FTO gene was selected as candidate genes.Haploview selected all labeled single nucleotide polymorphism.FTO genes rs1421085,rs1558902 and rs9939609 sites were selected for typing using time-flight mass spectrometry.The chi-square test was used to calculate whether the distribution of each genotype was in accordance with the Hardy-Weinberg balance,and the association with a single SNP was statistically analyzed using PLINK software.The difference of indexes before and after 16 weeks exercise intervention was analyzed by T-test of paired samples.Results: Initial value: After adding BMI,age and region as covariates,1)the maximal oxygen uptake test index: in male and female subjects,there was no association with the three sites.2)blood metabolism index: The rs1421085 polymorphism of the FTO gene was associated with HDL-C in female subjects.CC genotype HDL-C was the highest.The rs1558902 polymorphism of the FTO gene was associated with HDL-C in female subjects.AA genotype HDL-C was the highest.The rs9939609 polymorphism of the FTO gene was associated with HDL-C in female subjects.AA genotype HDL-C was the highest.Other indicators were not associated with three loci in both male and female subjects;16-week exercise fitness effect: Women in the exercise group had a significant decrease in body weight(P=0.001),BMI(P=0.001),percentage of fat content(P=0.004),significant increase in HDL-C(P=0.001),absolute value of VO2max(P=0.01),VE(P=0.002),CHOL(P=0.03),CO2(P=0.001)in the exercise group,and men in the exercise group had a significant increase in VO2max(P=0.03),VE(P=0.001),CO2(P=0.001),ATVE(P=0.01),HDL-C(P=0.006),CHOL(P=0.02).When age,region,BMI and initial values were added as covariates,there was no significant association between the maximum oxygen uptake index,body component test index and blood metabolism index in male and female subjects.Conclusion: 1)The rs1421085,rs1558902 and rs9939609 polymorphism of FTO gene can predict the concentration of HDL-C,the AA and CC genotype have higher HDL-C.2)FTO gene polymorphism could not predict fitness effects.
