The traditional medicinal plant Stellera chamaejasme L.is not only the "steppe poisonous grass" in urgent need of control,but also a "treasury" of natural compounds with abundant resources.Its active components have remarkable effects in insecticidal,anti-tumor and anti-HIV viruses.The preliminary test results showed that: the fractions 7and 8 in the extract of Stellera chamaejasme L.petroleum ether had the best therapeutic effect on TMV.Identified by HPLC-ESI-MS and found the same compound,Chamaejasme A and C,which Chamaejasme A accounted for 77.92%;and found that the extract has a certain promoting effect on the test plants.Therefore,in order to clarify the main mechanism of action of Chamaejasme A and C of the host to anti-TMV,this experiment used TMV-GFP as the test source,studied the optimal concentration of Chamaejasme A and C standard on TMV,and the host resistance defense response it causes.The test results are as follows:1.The activity of Chamaejasme A and C on TMV was determined by leaf plate method.Chamaejasme A showed inhibitory effect on the spread of infection of TMV at 20 ?g·m L-1,and Chamaejasme C had certain influence on the spread of virus at 40 ?g·m L-1.Transmission Electron Microscopy observation showed that the structure of virus in vitro was damaged by Chamaejasme A,causing them to break and partially degrade.2.After treatment with N.b at 20 ?g·m L-1,Chamaejasme A had an inhibitory effect on the early infection and spread of the virus.However,after treatment with N.b at 20 ?g·m L-1,Chamaejasme C promotes the entire replication and propagation of the virus and accelerates the systemic infection of the virus in the host.3.Chameaejasmin A promotes the expression of NPR1,PR1,and PR2 defense genes,induces the production of disease-related proteins,and improves host disease resistance;Chameaejasmin C has a certain effect on the early expression of PR1 and PR2 genes.In the signal transduction pathway mediated by Jasmonic acid,Chameaejasmin A and C have a certain promotion effect on the expression of MYC1 a gene,but there has any effect on the COI1 gene.In the Ethylene-mediated signal transduction pathway,each treatment group had a certain role in promoting the expression of ERF1 gene.4.After treatment of N.b plants with Chameaejasmin A and C,there has a certain effect on the activity of defensive enzymes in the plant and can induce the promotion of related defensive enzymes in plants.In addition,after treatment with Chameaejasmin A,the MDA content in the plant cells was significantly reduced,reducing the effect of membrane lipid peroxidation on the cells.After the treatment with Chameaejasmin C,the content increased sharply,and the damage to plant cell membrane structure was deepened.
