In recent years,studying on keratin genes,keratin associated protein genes,bone morphogenetic protein genes have found that the genes were involved in regulation of mammalian hair follicle cycle and hair growth,which were valuable genes in regulation hair growth.In view of these genetic studies was a correlation between a single gene and hair follicle.But it is not clear that a cell signaling pathway plays a role in the interaction among keratin,keratin associated protein and bone morphogenetic protein genes.DCN was one of the essential genes in the regulation of hair follicle growth.At present protein(FGF-5,IGF-1)and internal secretion(Melatonine,MT)played a role in the regulation of hair follicle development cycle.How to affect the relationship among protein,internal secretion and DCN gene is still not clear.Therefore,the motif of thesis studied on the mechanism of K26,KAP26.1,DCN and cell signaling pathway in mammalian skin fibroblasts,which was a newly direction for the growth developing of mammalian hair and laid the foundation for treatment of human diseases.The thesis was divided into two parts.The first part that studied on the effect of K26 and KAP26.1 gene over express on other Keratin associated protein,BMP-4 and BMPR-IB in mice skin fibroblasts,and KAP6.2,KAP7.1,KAP8.2,KAP11.1,BMP4,BMPR-IB were detected by Quantitative Real-time PCR in K26 or KAP26.1 over expression virus transfected mice skin fibroblasts in vitro,and explored relationship between K26 or KAP26.1 over expression and other genes.The second parts that the Liaoning cashmere goat skin fibroblasts were treated with MT,IGF-1 and FGF-5 in vitro by Quantitative Real-time PCR detected the relative level of DCN.Basing on the effect of skin fibroblasts treatment with MT and IGF-1 or MT and FGF-5,and detected the relative level of DCN goat skin fibroblasts in vitro.Result:(1)After K26 over expression,the relative level of BMP4 and KAP26.1 were up-regulation in mice skin fibroblasts.Meanwhile,after K26 over expression,the relative level of KAP6.2,KAP7.1,KAP8.2,KAP11.1 and BMPR-IB were down-regulation in vitro of mice skin fibroblasts.More were,when KAP26.1 over expression virus transfected mice skin fibroblasts,the relative level of KAP6.2,KAP7.1,KAP8.2,KAP11.1 were down-regulation in the mice skin fibroblasts.After the KAP26.1 was over expression,the relative levels of BMP4 and BMPR-IB were up-regulation.(2)Cashmere goat skin fibroblasts treatment with MT showed that low concentration MT promoted DCN expression and high concentration suppressed DCN expression.The skin fibroblasts treatment with FGF-5 showed that the higher the concentration of FGF-5,the more significantly of promotion DCN expression in 72h.The skin fibroblasts treatment with IGF-1 showed that IGF-1 significantly inhibited DCN expression.The DCN gene expression was down-regulati-on treatment with simultaneous of 0.02 g/L MT and 10-4 g/L FGF5 in 72 h.The relative level of DCN was treated with simultaneous of 10-5IGF-1 and 0.02 g/L MT in 72 h,which was down-regulation trend in skin fibroblasts.Conclusions:(1)The experimental data that the cooperative relationship between the K26 and KAP26.1.After the K26 or KAP26.1 gene over expression,the relative expression levels of KAP6.2,KAP7.1,KAP8.2,KAP11.1 showed a down-regulation trend,which involved in positive regulation in thicker of wool fibers,so that the wool fiber has an impact on quality.On the contrary,the K26 or KAP26.1 gene over expression increased the expression of BMP-4 and activated BMP signaling,leaded hair growth to be maintained in telogen.The K26 gene over expression eased the expression of BMPR-IB and inhibited the BMP signaling,which leaded hair growth to enter anagen from telogen.However,KAP26.1gene over expression increased the relative expression levels of BMPR-IB and activated the BMP signaling and leaded hair growth to be maintained in telogen.In summary,K26 and KAP 26.1 regulated KAP6.2,KAP7.1,KAP8.2,KAP11.1,BMPR-IB,BMP-4 expression,which showed that K26 and KAP26.1 were identical with that of the KAP6.2,KAP7.1,KAP8.2,KAP11.1,BMPR-IB,BMP-4 and participated in BMP signaling pathway,which was significance to regulate the development of mice hair.(2)The relative expression levels of DCN in cashmere goat skin fibroblasts treatment with FGF-5 and low concentrations MT were increased,which inhibited TGF-?signaling pathway in catagen,and then contributed to the growth of cashmere wool.On the contrary,the relative expression levels of DCN in cashmere goat skin fibroblasts treatment with IGF,simultaneous of IGF-1 and MT,simultaneous of FGF5 and MT,high-concentrations of MT were down-regulated,which activated TGF-?signaling pathway and made the hair follicle transformed from anagen to catagen,and then inhibited the growth of cashmere wool.
