Synthesis of phthalocyanines and naphthalocyanines for photodynamic therapy and photothermal therapy studies

Abstract/Summary:
A group of forty-seven new phthalocyanines and naphthalocyanines which were designed mostly for use in photodynamic therapy and photothermal therapy studies was synthesized and characterized. One was SiPc[OSi(CH3) 2C3H6Br]2, seven were of the type SiPc[OSi(CH 3)2C3H6N(CH3)2][OSi(CH 3)2C3H6R] where R contains a primary amine, biotin, rhodamine, acetate, alcohol, mesylate or sulfonate functionality, two were of the type SiPc(dib)4(OBu)8[OSi(CH3) 2C3H6R]2 where R is a tertiary amine or piperazine functionality, two were of the type SiPc(dib)4(OBu) 8[R]2 where R is an alkylsiloxy or cage siloxy group, one was H2Pc(dib)4(OiBu)8, and three were of the type SiPc(dib)4(OiBu)8[R]2 where R is a hydroxy, alkylsiloxy or cage siloxy group.; An additional one was H2Pc(OBu)8(CH2OC 3H6OC5H9O), five were of the type NiPc(OBu) 8(CH2OC3H6R) where R contains a pyrane, alcohol, mesylate, primary amine or fluorescein group (these were prepared by a lengthy but robust multi-step synthesis). Two were of the type H 2Pc(OBu)8(CH2OC3H6R) 4 where R is a pyrane or alcohol group, and six were of the type NiPc(CH 2OC3H6R)8 where R is one of two tertiary amine groups or one of four quaternary ammonium salt groups.; Two were of the type SiNc(OBu)8[OSi(CH3)2 C3H6R]2 where R is a tertiary amine or piperazine group, and one was SiNc(OBu)8[(OSi(CH3) 2)3CH3]2.; A pair was of the type H2Nc(OBu)8(CH2OC 3H6R) where R is a pyrane or alcohol function, and seven were of the type NiNc(OBu)8(CH2OC3H 6R) where R contains alcohol, mesylate, primary amine, fluorescein, tosylate, iodo or azide group (these also were prepared by a lengthy but robust multi-step synthesis). Two were of the type H2Nc(OBu)8(CH 2OC3H6R) where R is a pyrane or alcohol function, one was NiNc(OBu)8(CH2OC3H6OH), and two were of the type H2Nc(OBu)8(CH2R) 4 where R is a pyrane and alcohol group.; A discussion of the potential of these compounds for PDT and PTT studies is given. The rhodamine, biotin and sulfonate compounds have potential for formulation and drug targeting studies, the cage compounds for Type I/Type II PDT mechanism studies, the nickel phthalocyanine and naphthalocyanine fluorescein compounds for drug targeting studies, and the quaternary ammonium salt compounds for bacteriocide studies.
