The hyperspectral and polarization imaging technology,which combines the spectrum and polarization organically,provides a great source of data and credibility for each imaging field.The paper is on the basis of ordinary optical microscopy system,by increasing spectral split module and polarization modulation module,successfully realized the construction and application of hyperspectral microscopic polarization imaging system.The liquid crystal tunable filter is used to filter the incident light rapidly and realize the wavelength switching in milliseconds without vibration.The rotating polarization module is used to modulate polarization to get image information under different polarization angle.Mueller matrix is used to calculate the stokes parameter images of emergent optical vector coming out from the sample.Then the hidden status of structure of the normal light intensity image can be found in the detection.Using the continuous single-band image to extract the spectral curve of different pixel regions,we can distinguish the substance which can' t be distinguished in the direct image,and truly realize the integration between the map and spectrum.Through the construction of related devices,control of software and later data processing,the system is designed and implements automatic measurement and data analysis of high spectral microscopic polarization imaging system.We achieved expected results in the experiments of calcite crystal,hair fibers and pathological tissue,which confirmed the correctness of the theory and the feasibility of the system.The work of this paper has increased the dimension of information detection in the field of ordinary optical microscopy,and it can provide a reliable technical solution for criminal investigation and medical pathological diagnostic.
