Borderline personality disorder(BPD)is a common personality disorder caused by a large amount of biological and environmental factors.Accumulating evidence suggests that childhood maltreatment is a risk environmental factor in the development of BPD,but research on the genetic pathology of BPD is still in its early stages,and very little is known about the oxytocin receptor(OXTR)gene.The purpose of this study is to explore the relationship between childhood maltreatment,OXTR gene polymorphisms and their interaction on borderline personality.The study included 765 inmates with high risk from an adult male prison in Nanjing,Jiangsu Province.According to the BPD scores in Personality Diagnostic Questionnaire-4(PDQ-4)(BPD score?4),participants were divided into borderline personality group(BP group,n=342)and non-borderline personality group(non-BP group,n=441).PDQ-4,Childhood Trauma Questionnaire Short Form(CTQ-SF),Barratt Impulsiveness Scale-11(BIS-11)were administered to assess participants.DNA was extracted from 5 ml of venous blood in 765 high-risk inmates.Genotyping of the OXTR single-nucleotide polymorphisms(SNPs)applied multiple PCR technology and high-throughput sequencing technology.SPSS24.0 and Haploview v4.1 were used for statistical analyses.Main results are as follows:(1)There were significant differences between BP group and non-BP group in parental rearing pattern and parental relationship.The drinking and drug use history in BP group were higher than that in non-BP group.And the differences were statistically significant(p<0.05).(2)There were significant positive correlations between BPD scores and childhood maltreatment total scores and its subtypes(p<0.01).After excluding other types of abuse,only emotional abuse and emotional neglect were still positively correlated with BPD scores(p<0.05).Further regression analyses indicated that emotional abuse and emotional neglect could significantly positive predict BPD scores(R~2=0.106,p<0.001).(3)There were significant differences in the genotype and allele frequencies of rs53576 and rs237897 between the BP and non-BP groups(p<0.05),and the BP group had higher rs53576 and rs237987 AA homozygous and A allele frequencies than that in the non-BP group.(4)The interactions between OXTR-rs53576 and physical abuse and sexual abuse could significantly predict BPD scores(p<0.05),and the rs53576 GG homozygous carriers were more susceptible to physical abuse and sexual abuse.The findings suggest that childhood maltreatment,OXTR gene variations and the interaction between them are important mechanism for the development of BPD.The interaction between OXTR gene polymorphism and childhood maltreatment confirms different susceptibility hypothesis.
