Objective:In modern society,due to the accelerated pace of life,increased work pressure,the incidence of psychological fatigue has become increasingly high,patients often appear mental inconcentration,memory loss,poor mood,lack of work motivation and other manifestations,serious cases can cause Alzheimer's disease,tumor,etc.Therefore,this study focuses on the development of intervention measures for psychological fatigue,and discusses the mechanism of lavender essential oil to intervene psychological fatigue,so as to provide a new treatment method for clinical improvement of psychological fatigue.Methods:The theoretical studyTheoretical research is mainly carried out from the following aspects:1)Overview of the development of aromatherapy;2)mechanism of action and safety comparison of different routes of aromatherapy administration;3)Theoretical and physiological basic research of aromatherapy inhalation therapy in Traditional Chinese medicine.Literature researchSystematic evaluation of clinical research on lavender's improvement of fatigue;computer retrieval of clinical research on lavender's improvement of fatigue in CNKI,Wanfang Data,VIP,PubMed and Web of Science database;Chinese database retrieval keywords:"atigue" and "lavender";The English database searches for the terms "fatigue"and "lavender".The retrieved literature was rearranged,read,information extraction and quality evaluation were conducted for the identified included studies,and outcome indicators were compared.Experimental studyThe rats were randomly divided into blank group,model group,mint group and lavender group.In addition to the blank group,the other three groups underwent 72h complete sleep deprivation.After successful modeling,the blank group,the model group were fumigated with deionized water,and the peppermint combined and lavender group were fumigated with essential oil of 1%concentration for treatment.At the end of the experiment,the efficacy was evaluated by general conditions(mental state,weight)and behavioral tests(grip strength,open field,elevated cross maze).Then test by ELISA experiments in rats serum Ghrelin,Leptin and Orexin-A content,HE observed in the rat hippocampus and gastric mucosal injury,immunohistochemical observation rat hippocampus and stomach GHSR and OBR,A positive OX1R expression,Western Blot experiments GHSR detection in the rat hippocampus and stomach,the OBR,OX1R protein expression,rt-pcr detection and hippocampus in the stomach Ghrelin,Leptin and Orexin-a GHSR and its receptors,the OBR,OX1R mRNA expression.The biological mechanism of lavender essential oil improving mental fatigue was analyzed layer by layer.Results:According to theoretical research,aromatherapy has a profound theoretical basis of Traditional Chinese medicine.Through comparison,it is believed that inhaled therapy is more safe and effective,and more suitable for clinical promotion.The literature study found that among the 14 included clinical studies on lavender intervention in fatigue,lavender essential oil was used most frequently,with as many as 12 articles,and the other 2 articles used lavender scented tea and lavender cream respectively.Inhalation was the most frequently used method in terms of intake mode,involving 10 studies,3 of the other 4 were applied externally,and the remaining 1 was oral scented tea.Compared with the blank group,the gripping power of the model group decreased(P<0.01).Compared with the model group,the gripping power of the rats in the peppermint group and the lavender group increased(P<0.01,P<0.01).Compared with the peppermint group,the clutching strength of the lavender group was higher,with statistical difference(P<0.01).Compared with the blank group,the total distance of activity,the total number of space crossing,the number of upright position,and the upright position time in the model group decreased,with statistically significant differences(P<0.01,P<0.01,P<0.01,P<0.01).Compared with the model group,the total distance of activity,the total number of crossing cells,the number of upright position,and the upright position time increased in the menthol group,showing statistical differences(P<0.05,P<0.059,P<0.01,P<0.01).The total activity distance of the lavender group increased(P<0.05,P<0.05,P<0.05,P<0.05).There was no statistical difference in total activity distance between menthol group and lavender group.Compared with the blank group,the modification time of the model group increased(P<0.01).Compared with the model group,the modification time of the menthol group was reduced,but there was no statistical difference.Compared with the model group,the grooming time of rats in the lavender group was reduced,showing a statistically significant difference(P<0.05).Compared with the blank group,the percentage of open arm residence time in the model group decreased(P<0.05).Compared with the model group,the percentage of open arm residence time increased in the mint group and the lavender group(P<0.01,P<0.01).Hippocampal immunohistochemical positive substances expressed GHSR and OX1R,which were increased in the model group compared with the blank group(P<0.01,P<0.01).Compared with the model group,the expression of menthol group decreased(P>0.05,P<0.01),while that of lavender group decreased(P<0,05,P<0.01).Compared with the blank group,the OBR expression in the model group was decreased(P<0.01).Compared with the model group,the expressions of menthol group and lavender group were increased(P<0.05,P<0.01).The positive gastric immunohistochemical substances expressed GHSR and OX1R,which were increased in the model group compared with the blank group(P<0.01,P<0.01).Compared with the model group,the expression of menthol group decreased(P<0.01,P<0.01),while that of lavender group decreased(P<0.01,P<0.01).Compared with the blank group,the OBR expression in the model group was decreased(P<0.01).Compared with the model group,the expression of menthol group and lavender group increased(P<0.01,P<0.01).Western Blot showed that the OBR protein expression in the model group was lower than that in the blank group(P<0.01).Compared with the model group,the protein expression of menthol group and lavender group increased(P>0.05,P>0.05).RT-PCR showed that mRNA expression of Ghrelin in the hippocampus was decreased in the peppermint and lavender groups compared with the model group(P<0.01,P<0.01).Compared with the blank group,the expression of Leptin mRNA in the model group was decreased(P<0.05).Compared with the model group,OX1R mRNA expression in menthol group and lavender group was decreased(P<0.01,P<0.01).Compared with the model group,the mRNA expression of GHSR in the lavender group was decreased(P<0.05).Compared with the model group,the mRNA expression of Orexin-a in the mint and lavender groups decreased(P<0.01 and P<0.01).Conclusion:Lavender intervention fatigue best way is to essential oils inhaled treatment,improvement of multiplatform water environmental law(MMPM)72 hours continuous sleep deprivation can be a very good copy mental fatigue rat model of rat muscle tension descent,kuang field activity,increased anxiety,elevated the maze open arm activity to reduce mental fatigue performance.Lavender essential oil aromatherapy can improve these adverse states and treat psychological fatigue.The mechanism of the action of essential oils may be related to the regulation of the contents of ghrelin,leptin and orexin A.
