Gastric ulcer(GU)can be divided into stress gastric ulcer and chronic gastric ulcer.With the double pressure of physiology and psychology brought about by the rapid economic development in China,the incidence of stress gastric ulcers is also on the rise.The clinical manifestations are upper abdominal pain,acid swallowing,belching and other symptoms.In severe cases,it can cause gastric bleeding shock Wait.Some studies have pointed out that under stress,the balance of anti-inflammatory factors and pro-inflammatory factors is destroyed,which triggers an inflammatory reaction of the gastric mucosa and then leads to the occurrence of stress gastric ulcers.Changes in the structure of the intestinal flora can cause a variety of diseases.Under the stress state,the results and diversity of the intestinal flora are changed through the HPA axis.There are literature studies on chronic stress that can change the intestinal flora abundance and The diversity of flora,there are a large number of documents that acupuncture treatment can relieve the pain of patients with stress gastric ulcer and achieve a curative effect.There are few studies on the impact of stress gastric ulcers on the content of inflammatory factors and intestinal flora,so this experiment uses a modified version of restraint-immersion stress(RWIS)to model and observe acupuncture The changes of inflammatory factors and intestinal microflora in stress gastric ulcer model rats before and after treatment are based on the possible mechanism of acupuncture treatment of stress gastric ulcer.ObjectiveThrough acupuncture at "Zhongwan" and bilateral "Zusanli",observe the effects of acupuncture on inflammatory factors and intestinal flora in the serum of stress-introduced gastric ulcer(SGU)rats The mechanism of acupuncture treatment of SGU.MethodsTwenty-eight male SD rats were randomly divided into blank group,model group,acupuncture group,and drug group,with 7 rats in each group.Except for the blank group,the remaining three groups used the restraint-immersion method to prepare the SGU model.After modeling,on the second day,the blank group and the model group were bound for 20 minutes a day for 5 days;the acupuncture group acupuncture "Zhongwan" and bilateral "Zusanli",and left the needle for 20 minutes,once a day for 5 days;The drug group was given 2 mg/L omeprazole enteric-coated tablet solution 2 mL intragastrically,once a day for 5 consecutive days.The Guth method was used to calculate the gastric mucosal injury index.HE staining was used to observe the morphological changes of the rat gastric mucosa.16SrDNA sequencing method was used to detect changes in intestinal flora.Results(1)Rat gastric mucosal injury index:Compared with the blank group,the model group's gastric mucosal injury index increased significantly(P<0.01).Compared with the model group,the gastric mucosal injury index of rats in the acupuncture group and the drug group decreased significantly(P<0.01,P<0.05).Compared with the two treatment groups,the gastric mucosal injury index of the acupuncture group was lower than that of the drug group(P<0.05).(2)Morphological changes of rat gastric mucosa:Compared with the blank group,the model group had severe gastric mucosal damage,glandular disorder,severe hyperemia,and massive red blood cell exudation;compared with the model group,the acupuncture group rats Most of the gastric mucosa is intact,with a small amount of red blood cells exuding and some cells are necrotic;the gastric mucosa structure of the rats in the drug group is relatively complete,with a small amount of red blood cells exuding and cell necrosis.(3)IL-4,IL-6,TNF-? content in rat serum:Compared with the blank group,the serum IL-4 content of the model group was significantly reduced(P<0.01),IL-6,TNF-? content Compared with the model group,the levels of serum IL-4 in the acupuncture group and the drug group were significantly increased(P<0.01,P<0.05),IL-6,TNF-?
