| OBJECTIVE: It is to observe the healing effect of Huang's Orthopedic Formula 1 and Gu Long Capsule on rabbit radial bone defect and the change of transforming growth factor-? in the process of fracture healing.So as to explore the possible mechanism of Huang's Orthopedic Formula 1 and Gu Long Capsule in treating bone defects.To improve the reference data for the clinical treatment of bone defect and nonunion by Huang's Orthopedic Formula 1 and Gu Long Capsule.Experimental method: Thirty-two pure healthy New Zealand white rabbits were used to make a bone defect model with the right radius of 1.5cm.They were randomly divided into four groups(n = 8),namely group A was externally applied with Huangshi Orthopedic No.1 and oral Gulong Capsule,group B was externally applied with Huangshi Orthopedic No.1,group C was orally administered with Gulong Capsule,and group D was blank.The concentration of TGF-? in the blood of experimental animals in each group was detected on the day of modeling,2 weeks,4 weeks,6 weeks and 8 weeks after treatment.The fracture was photographed by X-ray at weeks 2,4,6 and 8.Results: The serum concentrations of TGF-? in each group were significantly different before surgery,and were basically similar without statistical significance(P> 0.05).Comparison of blood concentrations of TGF-? measured at 2 weeks,4 weeks,6 weeks,and 8 weeks after administration: The concentrations in groups A,B,and C at each time period were higher than those in group D,with significant differences(P <0.05);Compared with the TGF-? concentration of the B and C groups in each time period of the group A,the concentration of the A group was higher than that of the B and C groups,which was statistically significant(P <0.05);the TGF-? of the B and C groups There was no significant difference between the concentrations(P> 0.05).Comparison of TGF-? concentration at different time points in the same group: In the A,B,C,and D groups,the TGF-? concentration was significantly increased in the second week after administration compared with before surgery;the fourth,sixth,and eighth weeks after administration,The increase of TGF-? concentration in each group was not obvious and tended to be stable.X-ray manifestations: In group A,the fracture line and fracture end disappeared completely,the callus was connected with cortex,and the callus density was the same as the surrounding bone.The healing degree of group B and group C is worse than that of group A.The healing degree is similar.A small part of the fracture end is not filled with callus,and the callus density is higher.In group D,the amount of callus was very small,the fracture end was slightly hardened,and part of medullary cavity was closed.Statistical analysis of x-ray scores showed that the scores of groups a,b and c were higher than those of group d at 6W and 8W after operation,with significant difference(p < 0.05).The scores of each time period in group a were higher than those in groups b and c,with significant difference(p < 0.05).The scores of group b and group c in each time period were close,with no statistical difference,p > 0.05.The comparison of time points in the group showed that the scores of group A,B and C were positively correlated with time,while the scores of group D were close to each other at the 6th weekend and 8th weekend.Conclusion: Huang's Orthopedic Formula 1 and Gu Long Capsule can stimulate the body to secrete and activate TGF-? rapidly.The activated TGF-? can stimulate the proliferation and differentiation of osteoblasts and bone marrow mesenchymal stem cells,indirectly increase the number of osteoblasts and bone marrow mesenchymal stem cells,and promote fracture healing.At the same time,activated TGF-? can inhibit the activity of osteoclasts to reduce bone destruction.The simultaneous use of Huang's Orthopedic Formula 1 and Gu Long Capsule can stimulate the body to secrete and activate TGF-? faster,thus accelerating the proliferation and differentiation of various osteoblastic cells and promoting fracture healing. |
