Objective: To study the intervention effect of massage on skeletal muscle injury caused by gastrocnemius muscle in rats.By detecting the expression of Myf5,Myod and other proteins in MRFs after massage intervention,the therapeutic effect of zheng's manipulation on skeletal muscle injury caused by gastrocnemius muscle was discussed.Methods: 72 spf-grade SD rats aged 8 weeks were randomly divided into blank group(group C),model group(group E)and experimental group(group T).Group E and group T were subjected to centrifugal running-table modeling,while group C and group E were given no intervention.In the experimental group,zheng's massage method of "kneading" was administered immediately after modeling,12 h,24h and 48 h,respectively,to observe the changes in vital signs of rats.The histological changes of gastrocnemius were observed by HE staining and electron microscopy,the changes of serum ck-mm concentration,and the expression of Myf5 and Myod proteins in the gastrocnemius tissues of rats were detected by Western Blot.Results: 1.Summarized more than 500 clinical cases of in-person diagnosis by professor zheng huaixian,summarized the characteristics of "stress on muscles and bones","stress on massage treatment in the rehabilitation process","rational application of massage techniques",and "put forward the primary and secondary relationship between massage and medication" in soft tissue injury2.General observation: the rats in group C were in good condition,while the rats in the modeling group(group E and group T)had decreased exercise ability,irregular breathing,and some limbs were slightly swollen.After the modeling,the exercise ability recovered slightly,and the recovery of group T was better than that of group E.3.Histomorphological observation: the gastrocnemius fibers in group C were arranged neatly at each time without significant difference.The myofibrils in E0 and T0 groups were arranged in disorder,and some inflammatory cells were infiltrated.The infiltration of E12 and T12 inflammatory cells was increased.The disorder of E24 myofibrils increased,the inflammatory cells in T24 group decreased,and the disorder of some myofibrils restored.The inflammatory cells of E48 were still relatively obvious,and the damage was severe.The collagen fiber arrangement was basically restored in the T48 group,and the infiltration of inflammatory cells was significantly reduced.4.Observation of ultrastructure: the myofibrils in each phase of group C were arranged neatly and showed no significant difference.The myofibrils in E0 and T0 groups were disordered and some mitochondria were swollen.The mitochondrial swelling in E12 and T12 groups was significant and increased.E24 myofibril disorder was obvious,T24 mitochondria swelling was reduced compared with the previous,and partial myofibril arrangement was restored.Myofibrils in E48 group were dissolved,mitochondria were swollen,and myofibrils in T group were basically restored to normal arrangement.5.CK-MM content level: the serum ck-mm concentration level of rats in group E and group T reached the peak immediately after the modeling,and gradually decreased with time,and was basically close to the level of group C at 48 h,with a significant decrease in group T compared with group E.Compared with each group,the serum ck-mm concentration of rats in group E was significantly higher than that in group T,showing a very significant difference(P<0.05).6.Western Blot results:(1)Myf5 expression: compared with group C at the same time,the expression of Myf5 protein in the gastrocnemius muscle tissues of rats in group E was significantly increased except for the 0h time phase,which was of very significant statistical significance(P<0.01).Compared with group C at the same time,the expression of Myf5 protein in the gastrocnemius muscle tissues of rats in each phase of the T group was significantly increased,showing a very significant difference(P<0.05).Compared with group E at the same time,the expression of Myf5 protein in gastrocnemius muscle tissues of rats in each phase of group T was all increased,showing significant difference(P<0.05).(2)Myod expression: compared with that of group C,there was no significant change in Myod protein expression in gastrocnemius muscle tissues of rats in group E at 0h,which was not statistically significant(P>0.05).Myod protein in gastrocnemius muscle tissues of rats in other phases was increased,showing a significant difference(P<0.01).Compared with the group C,the expression of Myod protein in the gastrocnemius muscle tissues of rats in each phase of the T group was significantly increased(P<0.05).Compared with group E at the same time,Myod protein expression in gastrocnemius muscle tissues of rats in group T at 0h and 12 h was increased,showing significant difference(P<0.05),while Myod protein expression in gastrocnemius muscle tissues of rats in group T at 24 h and 48 h was increased,showing very significant difference(P<0.01).Conclusion:(1)the "kneading method" in zheng's manipulation has a positive effect on the repair of gastrocnemius muscle injury in rats.(2)Myf5 and Myod of myogenic factor family(MRFs)are involved in the process of skeletal muscle injury in rats,and the Wnt signaling pathway is one of the repair pathways of EIMD.(3)the "kneading method" in zheng's massage can significantly up-regulate the expression of Myf5 and Myod of myogenic factor family proteins in the rat model of calf muscle tissue injury,effectively promote the activation of Wnt signaling pathway,improve the expression of myogenic factor family,and thus play a role in the repair of motor skeletal muscle injury.
