Purpose:To investigate the effects of Tanshinone ?A and Salvianolic acid B on myocardial tissue in mice with coronary artery injury induced by Kawasaki Disease,and to explore the mechanism of its related effects,in order to provide a new idea for the treatment of myocardial injury caused by coronary artery injury of Kawasaki disease with traditional Chinese medicine.Material and method:Seventy two BALB / C mice were randomly divided into 6 groups: Control Group,Model Group,positive drug group,Tanshinone ?A + Salvianolic acid B group(1:1),Tanshinone ?A + Salvianolic acid B group(1:5),Tanshinone ?A + Salvianolic acid B group(5:1).Except for the blank control group,the other groups were modeled by intraperitoneal injection of 2.5 g / kg bovine serum albumin on day 1,3,5,7,9,and 11 respectively,while the blank control group was given 2.5 g / kg saline intraperitoneal injection to observe changes in mice activity,appetite and response.After modeling,the positive drug group was given the dose of 455 mg / kg for the first 3 days and 45.5mg / kg for the other 7 days,and the Chinese Medicine Group was given the dose of 40 mg / kg for 10 days,the Control Group and the model group were given the same amount of normal saline for 10 days.After 24 hours of the last administration,the samples were taken from mice,the blood from eyeball was detected by ELISA,the coronary artery tissue and myocardial tissue was stained by HE,and the pathological changes were confirmed.Results:1.Mice behavior: Tanshinone ?A combined with Salvianolic acid B after 10 days of treatment,the activity of mice returned to normal,increased diet and water,improve the quality of life.2.Myocardial histomorphological changes in mice: Tanshinone ?A combined with Salvianolic acid B could change the rupture of myocardial tissue in KD-CAL mice.Compared with model group,the myocardial tissue in Tanshinone ?A + Salvianolic acid B(1:5)group was almost normal,the structure is arranged in an orderly and compact way.3.Indicators of drug effectiveness:(1)The activity of CK serum of mice: Compared with the model group,Tanshinone ?A combined with Salvianolic acid B group decreased significantly(p< 0.05);Compared with the positive group,Tanshinone ?A + Salvianolic acid B group(1:5)decreased significantly(p< 0.05).(2)The activity of CK-MB serum of mice: Compared with the model group,Tanshinone ?A + Salvianolic acid B group(1:1),Tanshinone ?A +Salvianolic acid B group(1:5)decreased significantly(p< 0.05).(3)The activity of LDH serum of mice: Compared with the model group,Tanshinone ?A combined with Salvianolic acid B group decreased significantly(p < 0.05).(4)The activity of ?-HBDH serum of mice:Compared with the model group,Tanshinone ?A combined with Salvianolic acid B group decreased significantly(p< 0.05).(5)The activity of ASTserum of mice: Compared with the model group,Tanshinone ?A + Salvianolic acid B group(1:1),Tanshinone ?A + Salvianolic acid B group(1:5)decreased significantly(p< 0.05);Compared with the positive group,Tanshinone ?A + Salvianolic acid B group(1:5)decreased significantly(p < 0.05).4.Detection of related indexes of drug action mechanism:(1)Compared with the model group,the level of TNF-? in the Tanshinone ?A + Salvianolic acid B group(1:5)was lower(p < 0.05),and there was no significant difference between Tanshinone ?A and Salvianolic acid B group(p > 0.05).(2)Compared with the model group,the levels of IL-6 in thepositive group,Tanshinone ?A + Salvianolic acid B group(1:1),Tanshinone ?A + Salvianolic acid B group(1:5)were lower(p < 0.05);Compared with the positive group,the levels of IL-6 in the Tanshinone ?A + Salvianolic acid B group(1:1)and Tanshinone ?A + Salvianolic acid B group(5:1)were higher(p< 0.05).(3)Compared with the model group,the levels of IL-1? in the positive group,Tanshinone ?A + Salvianolic acid B group(1:5),Tanshinone ?A +Salvianolic acid B group(5:1)were lower(p< 0.05);Compared with the positive group,the levels of IL-1? in the Tanshinone ?A + Salvianolic acid B group(1:1)and Tanshinone ?A +Salvianolic acid B group(5:1)were higher(p< 0.05).Conclusion:1.Tanshinone ? A,a liposolubility effective part of salvia miltiorrhiza,combined with Salvianolic acid B,a water solubility effective part of salvia miltiorrhiza could inhibit myocardial tissue changes and reduce myocardial necrosis in mice with coronary artery injury induced by Kawasaki Disease.2.Tanshinone ?A combined with Salvianolic acid B at the ratio of 1:5 had the most significant inhibitory effect on myocardial injury.3.The mechanism of tanshinone ?A combined with salvianolic acid B in treating KD-CAL caused myocardial tissue damage may be related to inflammation-related cytokines TNF-?,IL-6 and IL-1?.
