Purpose:To explore the relationship between the composition of different intestinal flora and the syndrome type of disease,by analyzing the composition of intestinal flora in children with different syndromes of Henoch-Schonlein purpura(HSP).Material and method:The children diagnosed as HSP from July 1,2018 to December31,2019 in the outpatient clinic of the First Affiliated Hospital of Liaoning University of Traditional Chinese Medicine were divided into bleeding due to blood heat group(abbreviated as S group)and qi deficiency and blood stasis group(abbreviated as N group)according to the syndrome type.Healthy children of the same age were normal control group(abbreviated as H group).Using 16 SrDNA high-throughput sequencing technology and OTU clustering of intestinal flora in children's fecal samples,diversity analysis and difference analysis were carried out according to the clustering results.Results:1.Diversity analysis: Alpha diversity:Sobs index: S Group(135.17±37.77)? H Group(152.86±32.15),P value=0.38>0.05,N Group(140.57±45.57)and H Group(152.86±32.15),P value=0.57>0.05.Shannon index: S Group(2.89±0.64)? H Group(3.40±0.33),P value=0.09>0.05,N Group(2.98±0.38)? H Group(3.40±0.33),P value=0.04<0.05.Beta diversity: Through PCoA analysis,it was found that the intestinal microflora structure of each group was quite different,and ANOSIM analysis was carried out at the same time,the R value=0.21>0,the P value=0.005<0.05.The difference between groups is greater than the difference between groups,and grouping has practical significance.2.Difference analysis: The genera of S and H groups with statistical differences included Streptococcus(1.99% vs 0.31%),Lachnospiraceae?NK4A136?group(0.18%:0.83%)and GCA-900066575(0.002% vs 0.02%).The genera that may have statistical differences include Streptococcus(1.99% vs 0.31%).The genera with statistical difference in N group and H group included Eubacterium hallii group(0.75% vs 5.91%),Fusicatenibacter(0.33% vs2.98%),Adlercreutzia(0.03% vs 0.25%),Lachnospiraceae N C2004group(0.02% vs 0.09%),norank?f?Lachnospiraceae(0.40% vs 0.15%),Oscarillibacter(0.26% vs 0.05%).potentially statistically different genera included Anaerostipes(0.57% vs 2.56%).Conclusion:1.Diversity of intestinal flora in children with different syndromes HSP decreased in varying degrees.2.HSP the composition of intestinal flora of different syndromes may be quite different.bleeding due to blood heat syndrome of HSP main were mainly increased by pathogenic bacteria HSP such as Streptococcus?Klebsiella,deficiency of qi and blood stasis HSP were mainly reduced by beneficial bacteria such as Eubacterium hallii group?Fusicatenibacter?Anaerostipes.3.Rational use of traditional Chinese medicine to regulate intestinal flora may be of positive significance for the treatment of HSP.
