Objective:The purpose of this experiment is to study and compare the intestinal microflora structure and diversity of patients with Henoch-Schonlein Purpura(HSP)and healthy people,which to further understand the pathogenesis of HSP and provide help for the treatment.Methods:From January 2019 to October 2019,patients with HSP were collected from the inpatient department of Dermatology Department in Affiliated Hospital of North Sichuan Medical College,in which 28 patients were included in the study,30 cases were included in the healthy control group for comparison.The V3-V4 region genes of microflora of all fecal samples were amplified and sequenced by using 16S rRNA gene high-throughput sequencing technology,and the data analysis was conducted to compare the differences in intestinal flora between the two samples..Results:There was no significant difference in sex(?2=1.673,P=0.196),age(t=0.597,P=0.553)between the two groups(P>0.05).After quality control,1585663 effective sequences were obtained from all 58 cases of HSP patients and healthy controls,with an average of 27339.02±5531.69 for each sample.Based on the effective data,2067 OTUs were formed after custering,including 685 identical OTUs in both groups,780 OTUs unique to in group P and 602 OTUs unique to in group H.In the total samples,99.68%of the effective sequences could be annotated to four bacterial phylum with the highest abundance,i.e.Firmicutes(64.22%),Bacteroides(27.57%),Actinomycetes(4.93%)and Proteobacteria(2.97%).In group P,Firmicutes,Bacteroidetes,Actinobacteria and Proteobacteria accounted for 62.31%,30.00%,3.66%and 3.68%,respectively.In group H,the corresponding proportions were 66.10%,24.88%,6.20%and 2.56%respectively.Lefse analysis of LDA>2 points,which means there was a statistical difference between the two groups(P<0.05).There were Unspecified_prevotellaceae(LDA=4.58),Prevotella(LDA=4.58),Eubacterium(LDA=3.25)in group P.There were Unspecified_Lachnospiraceae(LDA=4.63),Roseburia(LDA=4.30),Haemophilus(LDA=4.04),Veillonella(LDA=3.90),Unspecified_Pasteurellales(LDA=3.88),Unspecified_pasteurellaceae(LDA=3.88),Acidaminococcus(LDA=3.64)in group H.Conclusion:The composition and abundance of intestinal flora of HSP patients were similar to that of healthy control group,but there were some significant different flora between the two groups.
