Purpose : The huoxueyibitang has a relatively obvious effect on clinical treatment of osteoarthritis.However,there is a lack of research on the mechanism of action of modern medicine.In this design experiment,by examining the activation state of signal pathways in the guinea pig knee joint tissues and blood,the regulation effect of self-made Huoxue Yibi Decoction on this pathway during treatment was explored.By exploring its possible mechanism of action,research can provide an experimental basis for the subsequent study of the preparation.Materlal and method: Choose 40 healthy 1-month-old female guinea pigs.These guinea pigs were randomly divided into 4 groups of 10 mice.The groups are:normal group,model group,traditional Chinese medicine group,and control group.The normal group directly collects and records.The other three groups of guinea pigs were raised to 5 months of age and used to establish OA spontaneous guinea pig models.The guinea pigs in the control group were fed by intragastric administration with glucosamine hydrochloride capsule solution.The experimental animals in the model group were given intragastric saline once a day.The Chinese medicine group was treated with zinihuoxueyibitang.These model animals were fed intragastrically for 30 days.After treatment,abdominal aortic surgery was performed to draw blood after anesthetizing coma guinea pigs with 20% uratan solution.Serum was collected by centrifugation at low temperature,and the contents of serum TLR4 and NF-k B were determined by enzyme-linked immunosorbent assay.At the same time,the soft tissue of the knee synovium of the guinea pig was intercepted and divided into two parts.After being infiltrated in physiological saline,some of them were stored in a-80 ? ultra-low temperature refrigerator.In another part,Western blotting was used to determine the expression of TLR4 and NF-k B key proteins in bone tissue.Results:1.Self-designed Huoxue Yibitang can significantly improve the general activity ofosteoarthritis guinea pigs.The weight gain of the treated guinea pigs is significantly different from that of other groups(P <0.05).2.Compared with normal guinea pigs,the levels of serum TLR4 and NF-?B in the guinea pigs of the model group were significantly increased(P <0.05).The difference was statistically significant,the levels of signal pathway factors were significantly increased,and a more obvious inflammatory response appeared.After treatment,the levels of serum TLR4 and NF-?B decreased(P <0.05).3.Compared with normal guinea pigs,the contents of TLR4 and NF-?B protein in synovial tissue of osteoarthritis guinea pigs were significantly increased(P <0.05).After treatment,the levels of TLR4 and NF-?B protein were down-regulated(P<0.05).4.Compared with the control group,the levels of TLR4 and NF-?B key proteins in guinea pig synovium treated with Huoxue Yibi Decoction were the highest,and the level of inflammatory inhibition was the highest.Compared with other groups,the difference was statistically significant(P <0.05).Conclusion:1.Zinihuoxueyibitang can control the level of inflammatory factors by controlling the activation path of TLR4 and NF-k B signaling pathways to achieve the effect of treating osteoarthritis.2.The therapeutic effect of zinihuoxueyibitang is more sensitive to the control of TLR4 and NF-k B signaling pathway compared with the control drug.Zinihuoxueyibitang has more advantages compared with the control drug.
