Purpose:To understand the current situation and existing problems of artificial breeding in the main producing areas of Rana temporaria chensinensis David in China,the draft technical regulations of producing and processing of Ranae Oviductus were formulated.Through the researches on Ranae Oviductus and the toads oviducts characteristics,microstructure,physical and chemical properties,chemical constituents,pharmacological actions of contrast,analysis of similarities and differences of the two,toxic ingredients of toads oviducts and to study the acute toxicity,provide experimental basis for toads oviducts resource utilization.Materials and methods:Examine objects and materials Liaoning and jilin have a certain scale of Rana temporaria chensinensis David breeding enterprises;There were 2 batches of oviduct samples from Rana temporaria chensinensis and 9 batches of oviduct samples from toad.Methods:1 Investigation on Artificial Domestication of Rana temporaria chensinensisField visit The field investigation was focused on the ecological environment of Rana temporaria chensinensis David and the incubation?breeding?stocking and wintering of Rana temporaria chensinensis David in artificial culture,as well as the production and processing of Ranae Oviductus.2 Comparative study of Ranae Oviductus and toads oviducts2.1 Comparative study of medicinal materials The dry samples of the two groups were observed and their characters were observed.The morphological similarities and differences of glandular cells were compared by using common light microscope.Dilatancy was determined according to the dilatancy determination method in the four parts of Chinese pharmacopoeia(general rule 2101).2.2 Comparative study of chemical composition The contents of water-soluble protein and total phospholipid were compared by spectrophotometry.HPLC method was used to compare the contents of 1-MH?amino acids?nucleosides?phospholipids?cholesterol and estradiol.2.3 Comparative study of pharmacodynamics The swimming model of mice was used and the results were analyzed statistically.Mice hypoxia experimental model was used and the results were statistically analyzed.2.4 Determination of toxic components and acute toxicity test of toads oviducts2.4.1 The content of Cinobufagin in toads oviducts was determined Cinobufagin was used as reference,the content of Cinobufagin in toads oviducts was determined by HPLC2.4.2 Acute toxicity test The acute toxicity test was conducted with reference to ?GB 15193.3-2003 food safety toxicology evaluation procedure?.Results:1 Investigation on Artificial Domestication of Rana temporaria chensinensisThe breeding situation is in the state of semi-artificial domestication.The technology of incubation and breeding is becoming more and more perfect.During the stocking period,it is in a state of no guardianship and no protection in the deep forest.At present,the methods of drying and peeling,freezing and fresh peeling are used.2 Comparative study of Ranae Oviductus and toads oviducts2.1 Comparative study of medicinal materials The dry samples of the two are different in appearance.The Ranae Oviductus is a block with a specific groove,and the toads oviducts is a chicken intestines with a curved layer of overlapping,both smell slightly fishy,but the former smell fishy,taste slightly sweet,the latter has a smell;The microstructure of glandular cells is similar,but the nuclei and fine lines of Ranae Oviductus cells are relatively more.Toads oviducts is less expansive than Ranae Oviductus.2.2 Chemical composition comparison2.2.1 The difference between the two groups was water-soluble protein and estradiol.2.2.2 The similar components of the two groups were: 1-MH?amino acids?nucleosides?phospholipids and cholesterol.2.3 Comparative study of pharmacodynamics The Ranae Oviductus could prolong the swimming time of the mice.There was no statistical difference between the toads oviducts group and the blank group,but there was a tendency to increase the exercise time of the mice.Ranae Oviductus could prolong the anoxia tolerance time of mice.There was no statistical difference between the toads oviducts group and the blank group,but it could significantly delay the occurrence of shortness of breath in mice.2.4 Determination of toxic components and acute toxicity test of toads oviducts2.4.1 The content of Cinobufagin in toads oviducts was determined Under certain experimental conditions,no Cinobufagin was detected2.4.2 Acute toxicity test After 14 days of continuous observation.Male and female mice showed normal activity,normal sleep,normal feeding and water intake,and no adverse reactions.Conclusion:1.The comprehensive survey shows that there are some problems in the production and processing of Ranae Oviductus.In order to promote the development of the industry,drew up ?Technical regulations for production and processing of medicinal materials of animal-derivative–RANAE OVIDUCTUS?.2.Similarities and differences:2.1 Similarities between Ranae Oviductus and toads oviducts: the structure of the glands of toads oviducts and Ranae Oviductus similar,chemical components of phospholipids? nucleosides?cholesterol?1-MH?amino acid content is not significantly different,the amino acid content in toads oviducts is more nutritious.2.2 Differences between Ranae Oviductus and toads oviducts: the contents of water-soluble protein and estradiol in chemical components were significantly different.In anti-fatigue and anti-stress pharmacodynamics,toads oviducts has no significant effect but has a trend of effect.3.Cinobufagin was not detected in toads oviducts,and there was no adverse reaction in acute toxicity test,so oral toads oviducts has certain safety.
