Purpose:To explore the interventional effect and mechanism of Baizhixue powder on ocular surface damage caused by experimental dry eye model of female C57 / BL6 mice.Material and method:In this experiment,a dry eye model of mice was induced by the intelligent dry environment system combined with the injection of scopolamine,and the experimental mice were divided into a blank control group(N),a model control group(M),and a Bai Zhi San powder administration group(B),Solvent control group(R),Cs A-positive drug control group(Y),7 mice in each group were tested for general health and ophthalmic items(periocular,conjunctiva,iris,cornea,optic disc and fundus vessels),tear fluid The secretory volume test(SIT),tear film rupture time test(BUT),and corneal fluorescein staining test(FL)ensure that the corneal barrier function of the 35 mice participating in the experiment is good,and the anterior segment and fundus conditions are normal.The dry eye model was established for a period of 14 days,and the administration was started on the first day of modeling.The administration period was 14 days.On the 14 th day,corneal epithelial damage detection,SIT,BUT detection were performed on mice in each group;Mouse eyeballs and eyelid appendage tissues were used to prepare frozen pathological sections;PAS glycogen staining was used to detect the density of goblet cells on conjunctival tissue of mice in each group;HE staining was used to detect the thickness of corneal epithelial cell layers in each group of mice Death method was used to detect the apoptosis of corneal and conjunctival epithelial cells in each group of mice;immunofluorescence staining was used to detect the infiltration of CD4~+ T cells in the conjunctival tissue of each group of mice.Results:After 14 days of oral administration of Tribulus Terrestris powder,compared with the model control group,the mice's tear secretion increased significantly;the tear film rupture time was prolonged;reduced sodium fluorescein retention was observed under cobalt blue light,and SIT,BUT,Compared with the model control group,the three FL indicators have significant differences(P<0.05),indicating that Baizhixue powder has a protective effect on corneal epithelial damage;HE staining results show that Baizhixue powder has a certain protective effect on corneal epithelial cell layers The thickness of the corneal epithelial cell layer in the administration group was significantly increased compared with the model control group,and the difference was statistically significant(P<0.05).The PAS glycogen staining results showed that Bai Zhi San powder reduced the lysis and loss of goblet cells,and the unit length of conjunctival tissue The number of upper goblet cells was significantly increased compared with the model control group(P<0.05);Tunel apoptosis experiment proved that Bai Zhi San powder can reduce the apoptosis of corneal epithelial cells and conjunctival epithelial cells in dry eye model mice,and compared with the model control group There was a significant difference(P<0.05);immunofluorescence staining revealed that the infiltration of CD4~+ T cells in the conjunctival tissue of mice after intragastric administration of Bai Tribulus powder was more significant.In the control group there was a significant reduction(P<0.05).Conclusion :The above research results show that the intelligent dry environment combined with the drug induction method can establish a fast and stable experimental mouse dry eye model.Using this animal dry eye model for 14 days after oral administration of Tribulus Terrestris powder,the ocular surface Epithelial damage was reduced,infiltration of CD4~+T cells in conjunctival tissue was reduced,tear secretion was increased,tear film stability was increased,goblet cell loss was reduced,and Bai Zhi San Powder could inhibit the apoptosis of corneal epithelial cells and conjunctival epithelial cells and Improve corneal barrier function.
