Purpose: An experimental study was conducted on the rat model of heart failure induced by isoproterenol by modified Linggui Zhugan decoction to observe the changes of heart function,LVEF,serum BNP,il-6 and other contents in the rats after medication,and the clinical efficacy of this formula was discussed,so as to explore a new idea and a new method for the clinical treatment of chronic heart failure.Material and method:Fifty SD male rats were randomly divided into the normal group(10 rats)and the heart failure group(40 rats)for modeling.the heart-failure group injected isoproterenol intraperitoneally,while the normal group injected normal saline.One in the heart-failure group died during modeling.LVEF was detected by echocardiography after one week of adaptive feeding followed administration.Modeling was indicated successful when LVEF was less than 50% in all heart-failure group rats whlie none in the normal group.Thirty-nine rats successfully modeled were divided into a modified Linggui Zhugan decoction group(10 rats),a captopril group(10 rats),a traditional Chinese and western medicine combinatorial treatment group(10 rats),and a model group(9 rats)in random number method.Rats in modified Linggui Zhugan decoction group were given the prepared solution 1.82 ml / d / per each,captopril solution 0.45 ml / d / per rat given in captopril group,in combinatorial treatment group a mixed solution of 1.82 ml of modified Linggui Zhugan decoction + 0.45 ml of captopril solution was given per rat daily.The normal group and the model group were given the same volume of normal saline daily.All groups were administered orally for 4 weeks.After the administration,rats’ general state was observed,cardiac ultrasound was performed and data was recorded.Then,animals were weighed,taken blood from the abdominal aorta,monitor serum BNP and IL-6,hearts were cut out and weighed,then cardiac body mass index was calculated.The heart tissue sectioned and HE stained to observe the changes of myocardium.Results:1.General state of rat :Compared with the model group,Three treatment groups can relieve the performance of fatigue,anorexia,and edema in heart failure rats to a certain extent.The best state was found in the combinatorial treatment group,and there was no significancely difference between the modified Linggui Zhugan decoction group and the captopril group.2.Cardiac ultrasound:According to LVEF,LVEDD,LVESD,LVPWd,LVPWs data,compared with the model group,three treatment groups can improve the rats’ cardiac function to a certain extent(P <0.01),combinatorial treatment group has the best effect(P <0.01).there was no significancely difference between the modified Linggui Zhugan decoction group and the captopril group(P >0.05).3.Cardiac mass index of rats : Compared with the model group,the heart weight index of the rats in three treatment groups were significantly improved(P <0.01).There was no significant difference between the three groups(P> 0.05).4.BNP levels: Compared with the model group,the BNP levels in three treatment groups were significantly reduced(P <0.01);Combinatorial treatment group has the best effect,and the captopril group is superior to the modified Linggui Zhugan decoction group(P <0.01).5.IL-6 levels: Compared with the model group,three treatment groups IL-6 levels were significantly decreased(P <0.01);Combinatorial treatment group was better than other two groups(P <0.01),and the captopril group is superior to the modified Linggui Zhugan decoction group(P <0.05).6.Myocardial HE staining was confirmed microscopically: Compared to the normal group,the other four groups all had varying degrees of damage,the damage degree order: the model group > the modified Linggui Zhugan decoction group and captopril group> the traditional Chinese and western medicine combinatorial treatment group.Conclusion:1.Modified Linggui Zhugan decoction can effectively improve the general state of rats,reduce the heart weight index and the levels of BNP and IL-6 in serum,and the effect is similar to captopril,but the best state was found in the combinatorial treatment group.2.Modified Linggui Zhugan decoction can improve cardiac LVEF,effectively improve cardiac function.3.Modified Linggui Zhugan decoction can reduce the myocardial cell injury in rats with chronic heart failure and effectively treat chronic heart failure.
