Objective:Studying the different expression levels of NF-?B?IL-1?and IL-18 in patients who suffer from Granulomatous Mastitis(GM)and breast fibroadenoma tumor,testing the the expression levels of IL-1?and IL-18 in the experimental group and control group before the treatment and after treatment in the patient of GM,to discuss the influence of the expression of IL-1?and IL-18 treating by Traditional Chinese medicine with GM.Methods:Preliminary experiments:We randomly select paraffin-embedded tissue which include 10 normal tissue adjacent to breast fibroadenoma tumor and 10 diagnosed with GM from our hospital's Breast inpatient department from April 2019 to September 2019.Then,we use IHC to measure the levels of NF-?B?IL-1?and IL-18.Meanwhile,we randomly select fresh tissue of 20 patients who meet the inclusion criteria in our hospital's Breast surgery from April 2019 to November 2019.A 5mm~3fresh tissue is cut from 10 patients with fibroadenoma of the breast and 10patients with granulomatous mastitis.The expression levels of IL-1?and IL-18 in the two groups are detected by ELISA,and the expression level of NF-?B are detected by RT-PCR.According to the results of the preliminary test,selecting the appropriate test index and test method for the formal experiment.Formal experiment:80 GM patients are randomly divided into two groups with 40 patients in each group from February 2019 to January2020.The method of the experimental group adopt Oral Chinese medicine+external treatment+surgical treatment,the control group adopt external treatment+surgical treatment.The course of treatment is 3months.In the enrolled patients,breast lesions are first harvested in abscess incision and drainage as before treatment specimens.After 3 months of treatment,the mass are limited,breast lesions are harvested at lumpectomy as after treatment specimens.The expression levels of IL-1?and IL-18 are detected by ELISA.Results:In the preliminary experiment,the expression levels of IL-1?and IL-18 in the normal tissue adjacent to breast fibroadenoma tumor and the granuloma group which are detected by ELISA are obviously different(P<0.01).There is undifferentiated expression of NF-?B between the two groups which is detected by RT-PCR(P>0.05).There are undifferentiated expression of NF-?B?IL-1?and IL-18 in paraffin-embedded tissue of the two groups which are detected by IHC(P>0.05).In the formal experiment,There are undifferentiated expression of IL-1?and IL-18 in the experimental group and control group before the treatment(P>0.05),The expression of IL-1?and IL-18 in the two groups after treatment are different,the expression level of IL-1?in experimental group is lower than the control group(P<0.05),the expression level of IL-18 in experimental group is significantly lower than control group(P<0.01),Comparing two group before and after treatment,the expression level of IL-1?and IL-18 are significant different(P<0.01).Conclusion:The expression of IL-1?and IL-18 are higher in the granuloma group.The expression of NF-?B does not increase in GM patients.Oral Chinese medicine+external treatment and external treatment can reduce the expression of IL-1?and IL-18(P<0.05,P<0.01),The results show that the combination of Chinese medicine+external treatment is superior to the single external treatment in down-regulating the expression of IL-1?and IL-18.
