Objective:The animal model of coronary heart disease with blood stasis syndrome was established based on the theory of "the heart and the small intestine sharing a paired relationship".The relationship between coronary heart disease with blood stasis syndrome and intestinal flora was discussed through the intervention of Yangxin Tongmai prescription,a famous Chinese medicine prescription.The effect of fecal bacteria transplantation was studied on intestinal microflora in rats with coronary heart disease with blood stasis syndrome.Methods:1.Effects of Yangxin Tongmai prescription on intestinal flora in rat model of coronary heart disease with blood stasis syndrome: The rat model of blood stasis syndrome of coronary heart disease was established by diet and drug induction.coronary heart disease after the success of the building,through the collection of rat serum drug intervention in two weeks,blood vessels,heart,small intestine biochemical tests,pathological examination and get the rat feces 16 s RNA sequencing,another rat feces then preparation of fecal bacteria mixture follow-up experiments should be carried out.2.Fecal bacteria transplantation for coronary heart disease(CHD)of rat of blood stasis syndrome animal model of the influence of intestinal flora: using high fat feed and oral lavage vitamin D3 with intraperitoneal injection method of isopropyl adrenaline duplicate rat model of blood stasis syndrome,coronary heart disease after the success of the building,oral lavage in different group rat feces bacterium mixture,two weeks after the intervention,in rat serum,blood vessels,heart,small intestine biochemical tests,pathological examination and get the rat feces to 16 s RNA sequencing.Result:1.Builded successful rats,diet water decreased,form the colour yellow withered,depressed,or neighbors huddled,lipid indexes except HDL,in the other were higher than in normal control group,the difference statistically significant(P < 0.01),ECG lead ? J point up or down,flat or inverted T wave.2.rats by drug intervention after the treatment,blood fat index TCHO,TG,LDL-C were reduced,HDL-C,statistically significant difference(P < 0.05 or P < 0.01).3.Pathological observation showed that the myocardial tissue cells in the model control group were incomplete with obvious inflammatory response and vascular wall stenosis.In the drug intervention group,the myocardial tissue cells in the medium dose group of Yangxin Tongmai prescription were basically complete,making it difficult to observe inflammatory response.In the model control group,the intestinal tissues were damaged and incomplete,with inflammatory infiltration and necrosis.In the drug intervention group,the intestinal tissues in the medium dose of Yangxin Tongmai prescription were basically intact,and inflammatory infiltration and necrosis could not be observed.4.Intestinal flora was extracted for high-throughput sequencing analysis of16 s RNA.In the model group and the healthy group,Bacteroidetes and Firmicutes had the highest abundance,accounting for 42.34% and 59.16% of Bacteroidetes and44.21% and 31.53% of Firmicutes,respectively.Proteobacteria,Fusobacteria and Actinobacteria have higher abundance.In addition,the differences between the two groups were as follows: the bacteroidetes in the atherosclerosis model group were significantly lower than those in the healthy group(P=0.014 < 0.05).The firmicutes in the model group were significantly higher than those in the healthy group(P=0.007 <0.01).The results of diversity analysis showed that there was no significant difference in bacterial population diversity between the two groups.OTUs comparison results showed that the pathogenic bacteria of Veillonella,Veillonelladispar,Scardovia and Mogibacterium increased significantly in the atherosclerosis group,while the beneficial bacteria of bacteroidetes such as Akkemansia,Turicibacteraceae and Bifidobacterium decreased significantly.5.The intervention of Yangxin Tongmai prescription could reduce TCHO,TG and LDL-C indexes and increase HDL-C indexes.The difference between the two groups was statistically significant(P < 0.05 or P < 0.01).6.After the intervention treatment of Yangxin Tongmai prescription,the intestinal flora in the middle and low groups of Yangxin Tongmai prescription recovered close to the normal group,compared with the coronary heart disease and blood stasis syndrome group and atorvastatin calcium group,the number of E There was statistical significance(P < 0.01);the number of lactobacilli and bifidobacteria in Yangxin Tongmai prescription group was higher than that in coronary heart disease and bloodstasis syndrome group and atorvastatin calcium group,the recovery was faster,the difference was statistically significant(P < 0.05 or P < 0.01).7.According to the results of fecal bacteria transplantation,the levels of beneficial bacteria in intestinal flora of rats with coronary heart disease with blood stasis syndrome who took the mixture of fecal bacteria in the normal control group and the drug intervention group increased significantly(P < 0.01).Conclusions: 1.The occurrence and development of blood stasis syndrome with coronary heart disease were affected by the disorder of intestinal flora in rats: the blood stasis syndrome with coronary heart disease may have mutual influence with the imbalance of intestinal flora,and the abundance of Bacteroides and pachytenella in the blood stasis syndrome with coronary heart disease group was significantly different from that in the normal control group.2.Yangxin Tongmai prescription can effectively treat rats with coronary heart disease with blood stasis syndrome,play a protective role in myocardial tissue and intestinal tissue,and improve blood lipid and hemorheology index.3.Yangxin Tongmai prescription was beneficial to the recovery of lactobacillus,bifidobacterium and akman bacteria in the intestinal flora of rats with coronary heart disease with blood stasis syndrome,and to the inhibition of pathogenic bacteria including escherichia coli,Veillonella,Veillonelladispar,Scardovia and Mogibacterium..It may be one of the mechanisms of Yangxin Tongmai prescription to prevent and cure coronary heart disease.The regulating effect of traditional Chinese medicine on intestinal microecology can be realized through a variety of ways,and the balance of intestinal microecology can be regulated only by nourishing the heart and clearing the veins,so as to achieve the purpose of treating rats with coronary heart disease with blood stasis syndrome.
