Objective:To observe the effect of moxibustion and acupuncture on the expression of IL-17 A,IL-22 and serum IFN-? in rats with Crohn's disease,and to explore the immune mechanism of moxibustion and acupuncture in relieving Crohn's disease.Methods: 32 SD rats were randomly divided into 4 groups,8 in each group,normal group,model group,model + moxibustion group,model + acupuncture group.TNBS was used to prepare Crohn's disease model rats.Moxibustion or acupuncture intervened on Tianshu point and Shangjuxu point for 15 minutes at the same time.Observe and record: behavior,gross morphology of colon,histomorphology after he staining,WB detection of IL-17 A,IL-22 and ELISA detection of IFN-?.Result:1.Behavior: compared with the normal group,the model group: hair thinning and crispy,part erect,prone to lying,anorexia and emaciation,significant weight loss,crouching abdomen and arched back,a large number of pale yellow paste stool around the anus,slow response to external stimulation,some rats have mucopurulent stool and so on.Compared with the model group,the behavior of model + moxibustion group and model + acupuncture group were improved.2.Morphology under visual inspection: compared with the normal group,the model group: irregular intestinal wall morphology,thickened intestinal wall,intestinal wall adhesion,linear ulcers of different sizes on the surface of mucosa,hyperemia and swelling,erosion,bleeding,high edema of intestinal mucosa,typical cobblestone like changes in the mucosa,such as polypoid,irregular thickening of blood vessel wall.Compared with the model group,the colon morphology of model + moxibustion group and model + acupuncture group was improved.3.Histology under light microscope: compared with the normal group,the model group: intestinal mucosa epithelial loss,irregular arrangement of glands,obvious thickening of submucosa,vascular proliferation,more monocytes and lymphocytes infiltration can be seen in the mucosa and submucosa,lymphoid tissue proliferation,slit like ulcer can be seen in the mucus membrane,part of the ulcer can reach the intestinal wall muscle layer,occasionally granuloma formation.And fibrotic scars.Compared with the model group,the tissue morphology of model + moxibustion group and model + acupuncture group was improved.4.The changes of IL-17 A,IL-22 and IFN-? were as follows:(1)compared with the normal group,the expression of IL-17 A,IL-22 and IFN-? in the model group increased significantly(P < 0.05).(2)Compared with the model group,the expression of IL-17 A and IFN-? in the model + moxibustion group decreased significantly(P < 0.05),and there was no statistical difference in the change of IL-22;there was no statistical difference in the change of IL-17 A,IL-22 and IFN-? in the model + acupuncture group.(3)Compared with the model + moxibustion group,the expression of IL-17 A and IFN-? in the model + acupuncture group increased significantly(P < 0.05),and there was no significant difference in the change of IL-22.Conclusion: 1.Moxibustion and acupuncture can improve the hair quality,mental state,reaction,food consumption,fecal properties and colonic mucosa inflammation of Crohn's disease model rats.2.Moxibustion and acupuncture may improve the immune status of colon mucosa by inhibiting the expression of IFN-? and IL-17 A,and then play a role in treating Crohn's disease.
