| Objective:To observe the effect of acupuncture intervention on neurological function of ischemic stroke score of rats and the expression of Bcl-2 and Bax,to explore the effectiveness of early intervention of acupuncture treatment of ischemic stroke in rats,to provide experimental evidence for the prevention and treatment of ischemic stroke for standardized clinical application of acupuncture.Methods:In this study,160 SD rats were randomly divided into sham operation group,model group,acupuncture group 2 hours and 168 hours of acupuncture group four groups,40 rats in each group,the rats according to 1 days,3 days,7 days,14 days and four observation points is divided into various sub groups in each group,10 rats were randomly assigned to.Preparation of focal permanent middle cerebral artery occlusion model(pMCAO)in rats by intraluminal suture.After modeling,2 hours in acupuncture group rats after 168 hours respectively to the acupuncture treatment,the pathological side Baihui through Qubin point,Fengchi Points,limbs of Neiguan and Zusanli,for 30min,during the lifting thrusting twirling 3 times,1 min each time,every day 1 times,continuous 14d,sham operation group,model group rats every day without acupuncture in the acupuncture group at the same time,but to crawl and bundled and fixed time and conditions with acupuncture group.At the four time point,the four groups were given neurological function score,decapitated rats were sacrificed,and the expression of Bcl-2,Bax and protein were detected by immunohistochemistry.All data were analyzed by statistical software SPSS 13.0 software.Result:1.Neurological function score(1)Compared with the sham operation group,sham group score(18 points)were not fall in the model group,acupuncture group 2 hours and 168 hours in the acupuncture group 1 days,3 days,7 days,14 days four time points of the 95%confidence interval,the difference was statistically significant;(2)Compared with the model group,the neurological score of 2H acupuncture group was higher than that of the former at the four time points,and the difference was statistically significant(P<0.01);(3)Compared with the model group,168h acupuncture group,in the model after completion of 1 days,3 days,7 days,nerve function improvement is not obvious,the difference was not statistically significant(P>0.05),on the fourteenth day after modeling when the nerve function improved significantly,the difference was statistically significant(P<0.01);(4)Compared with 168h acupuncture group,2h acupuncture group at the time of the four points,neurological function score were significantly improved,the difference was statistically significant(P<0.01);2.Expression of Bcl-2 and Bax protein(1)Compared with the sham operation group,model group and acupuncture group(two 2h,168h acupuncture group)the expression of Bcl-2 protein and Bax protein expression increased significantly,and the rats in the completion of the first day,the expression reached the peak,then decreased,the difference was statistically significant(P<0.001,P<0.01);(2)Compared with the model group,the expression of Bcl-2 protein was increased and the expression of Bax protein was decreased in the 2H acupuncture group at the time of the,the difference was statistically significant(P<0.001);(3)Compared with the model group,the expression of Bcl-2 and Bax protein in the 168h acupuncture group was not significantly changed at the time of the completion of the model on the 1 day,the 3 day and the 7 day after,and the difference was not statistically significant(P>0.05).(4)Compared with 168h group,the Bcl-2 protein in the 2H group was significantly increased at the time of the four groups,and the expression of Bax protein decreased significantly(P<0.001)Conclusion:1.Acupuncture intervention can significantly improve the neurological function score of cerebral ischemia model rats,so as to improve their motor,sensory and coordination functions.2.Acupuncture intervention can reduce the expression of Bax protein in brain tissue of rats with cerebral ischemia,increase the expression of Bcl-2 protein,inhibit the apoptosis of nerve cells,and play a neuroprotective role.3.Early intervention of acupuncture can improve the neurological function of cerebral ischemia rats,reduce the expression of Bax protein in brain tissue of cerebral ischemia model rats,and increase the expression of Bcl-2 protein. |
