Purpose:This experiment selects the loperamide hydrochloride suspension liquid preparation of functional constipation(FC)in rats model,the rats model of constipation were treated by applying the traditional Chinese medicine on both sides of "tianshu" point,by observing the defecation function changes in functional constipation model rats,morphological change of colon was observed under light mircroscope,detecting the FC variation in content of rat serum GAS,MTL and colonic myenteric plexus the expression of VIP and SP,to investigate the effect of acupoint application of traditional Chinese medicine on pathological changes of colonic tissue gastrointestinal hormones and neurotransmitters in rats with functional constipation.Material and method : Forty SD male rats were randomly divided into the blank group,the model blank group,the acupoint application group and the positive drug group after a week of adaptive feeding,with 10 rats in each group.The model blank group,the acupoint application group and the positive drug group were given loperamide hydrochloride suspension(0.5mg/ml)daily by gavage,3mg·kg-1·d-1,the blank group was given normal saline of the same volume.After a week of continuous molding,water could not be prevented from fasting for 24 h,and 2ml of activated carbon suspension was given by gavage.The timing was started after the completion of activated carbon suspension gavage,and the first black defecate excretion time,fecal grain number,and fecal quality within 6h were recorded,and moisture content was calculated according to the fecal quality.Compared with the blank group,the difference the was statistically significant(P<0.01),indicating that the modeling was successful.After the successful modeling,the blank group and the model blank group were given blank application on both sides of "tianshu" acupoints,the acupoint application group was given the intervention of Chinese medicine application on both sides of "tianshu" acupoints,the medicine contains Rheum officinale、Magnaolia officinalis、Glauber salt、Cassia seed、Barbados aloe、Borneol,gring into fine powder in the ratio of 1︰2∶1∶1∶1∶1,add ginger juice to make it into paste,take 0.5cm×0.5cm size medicine block at a time and put iton 3cm×3cm size medical nonwoven cloth for sticking.The positive drug group was given the intervention of moxapilide suspension(0.15mg/ml)by gavage,1.58mg·kg-1·d-1.The acupoint application group was treated for 6 hours a day,and the positive drug group was treated once a day,total four weeks.After the end of the last intervention,each group of rats was placed in a single metabolic cage,fasting without inhibiting water for 24 h,and then gavage with activated carbon suspension.The timing was started from the end of gavage with activated carbon suspension.The first black defecation time,the number of feces,and water content within 6h were recorded,to observe the changes in the defecation function.The morphological changes of colon tissues of rats in each group were observed by HE staining under light microscope.The contents of GAS and MTL in rat serum were determined by enzyme linked immunosorbent assay(ELISA),the expression levels of VIP and SP in the colonic muscle plexus were detected by immunohistochemistry,and the average optical density of the positive expression were calculated by morphological image analysis system.Results:1.Compared with the blank group,the first black defecation of the model blank group was prolonged,the number of feces was small,and the fecal moisture content was low(P<0.01).Compared with the model blank group,the first black stool excretion time was shortened,fecal grains were increased,and fecal moisture content was increased in the acupoint application group and the positive drug group(P<0.01).2.Through observation of pathological sections of rats in each group,the lesion degree of acupoint application group and positive drug group was less than that of model group.3.Compared with the blank group,the content of GAS and MTL in the model blank group decreased significantly(P<0.01).Compared with the model blank group,the content of GAS and MTL in serum of acupoint application group and positive drug group was significantly increased,and the difference was statistically significant(P<0.01).4.Compared with the blank group,the average optical density values of VIP and SP in the colonic muscle plexus of the model blank group all decreased(P<0.01).Compared with the model blank group,the average optical density of VIP and SP in the colonic muscle plexus inthe acupoint application group and the positive drug group was significantly increased,and the difference was statistically significant(P<0.05).Conclusion:1.The application of traditional Chinese medicine on both sides of "tianshu" acupoint can shorten the time of first black defecation,increase the number of stool grains,improve the moisture content of the stool,and improve the function constipation of the rats.2.The application of traditional Chinese medicine on both sides of "tianshu" acupoint can effectively improve the pathological status of functional constipation.3.The application of traditional Chinese medicine on both sides of "tianshu" acupoint can improve constipation by increasing the content of GAS and MTL in the serum of rats with functional constipation,and regulating gastrointestinal hormones.4.The application of traditional Chinese medicine on both sides of "tianshu" point can improve the expression level of VIP and SP neurotransmitters in colonic muscle plexus of rats with functional constipation,promote the recovery of colonic neurotransmitter function,enhance colonic transit movement,and improve constipation...
