Purpose:This experiment intends to establish a model of type 2 diabetes mellitus with qi and yin deficiency by means of diet-induced combination of chemical reagents and traditional Chinese medicine.And observed the expression of circulating fetuin A(FetuinA)and circulating fetuin B(FetuinB)in the model rats after the intervention of traditional Chinese medicine compound Yitangkang,and analyzed the glucose and lipid metabolism indicators of each group,thus the compound benefit of traditional Chinese medicine.The effectiveness of Ginkang is evaluated,and new ideas for the prevention and treatment of type 2 diabetes are provided.Material and method:1.Preparation of type 2 diabetic rats with qi and yin deficiency Healthy normal male Wistar rats were selected,8 weeks old,totaling 70(Liaoning Changsheng Biotechnology Co.,Ltd.).Rats were given free access to drinking water and fed for 7 days.Rats that were well-adapted and free to move were included in the formal experiment.Rats were randomly divided into 10 groups in the initial group and 70 in the STZ group.Rats in the STZ group were fed with high-fat and high-sugar diet,and the blank group was fed with normal diet for 4 weeks.The STZ group rats were then injected with intraperitoneal streptozoon(STZ).The normal group was given the same amount of buffer injection,and it was determined that the random blood glucose was greater than 16.7 mmol/L for the successful modeling of type 2 diabetes.The model rats were randomly divided into T2 DM group and T2 DM qi and yin deficiency group.Rats in T2 DM Qiyin deficiency group were over-administered with Chinese medicine Qingpi,Fuzi granules and rats in type 2diabetes group for the same amount of normal saline for 4 weeks.The general condition of the mice was observed and the modeling was successful.The blank group does not do anything.2.Determination of circulating Fetuin A and circulating Fetuin B levels in different groups.Serum circulating Fetuin A and circulating Fetuin B levels were determined in different ways by Elisa method,and observe their association with glucose and lipid metabolism indicators.3.Intervention of Chinese medicine compound YiTangKangThirty rats of T2 DM qi and yin deficiency group were randomly divided into 10 groups of qi and yin deficiency model and 20 rats in the experimental group.Rats in the experimental group were randomly divided into Yixukang treatment group 10 and metformin group 10,Yiyikang treatment group and western medicine control group were converted into equivalent doses of rats,and Qi and Yin deficiency model group was given.The same amount of normal saline for a total of 8 weeks.Results:1.After STZ injection,the blood glucose of the STZ group was significantly changed compared with the rats in the blank group,and all of them were greater than 16.7 mmol/L.After the model of qi and yin deficiency,the rats in the T2 DM qi and yin deficiency group showed diet,drinking water,increased urine output,body weight loss,tail defecation,urination,etc.,and the stool smell increased,fatigue,laziness,joy,lying or Piled up lying,a few irritability,easy to panic,dark brown,dull,thick,low-grade,thin,and non-mosquito,such as yin and yin.It is consistent with the evaluation criteria for the type 2 diabetes mellitus with Qi and Yin deficiency.2.After the end of the experiment,the uric acid(UA)and high-density lipoprotein(HDL-C)in the T2 DM group were higher than those in the T2 DM qi-yin deficiency group,total cholesterol(TC),triglyceride(TG),and fasting blood glucose.FBG)and low-density lipoprotein(LDL-C)were lower than those of Qi-Yin deficiency model(P>0.05),but the expressions of FetuinA and FetuinB were higher than those in T2 DM group(P<0.05).),the two indicators have a significant impact on type 2 diabetes qi and yin deficiency syndrome.3.After 8 weeks of intervention with Chinese herbal compound Yitangkang,FetuinA expression was decreased and FetuinB expression was increased(P<0.05).There was no significant change in metformin treatment group(P>0.05).The general condition and FBG ofYitangkang treatment group rats.Fasting insulin(FINS),TG,TC,HDL-C,LDL-C),UA and other glycolipid metabolism indicators were significantly improved,and except FBG,were superior to metformin group.Conclusion:1.By feeding high-fat and high-sugar diet,combined with STZ intraperitoneal injection,intragastric administration of gas-stasis and yin-like traditional Chinese medicine,the rat model of qi-yin deficiency syndrome can be successfully reproduced.2.The expression of FetuinA and FetuinB has a significant impact on type 2 diabetes mellitus with qi and yin deficiency syndrome,and can be used as an objective indicator to identify type 2 diabetes mellitus with both qi and yin deficiency and non-qi and yin deficiency.3.Improve the expression of Fetuin family-related protein to treat type 2 diabetes mellitus with Qi and Yin deficiency.It is one of the mechanisms of traditional Chinese medicine compound Yitangkang.The curative effect is true,and the overall effect is better than the western medicine metformin.
