| Purpose:To study the mechanism and treatment effectiveness of reflux esophagitis by using Shuganlidanhewei decoction through evaluated the expression of esophageal tissue and histopathology,lower esophageal pH and IL-8 values of esophageal mucosa in rat model.Finally compare the treatment results between Esomeprazole and Shuganlidanhewei.Material and method:160 SPF,SD rats weight;200-220 g were used.The rats were divided into western medicine,Chinese medicine,model and sham operation groups.Each group has 40 rats.Reflux esophagitis model was prepared by pylorus half ligation.Dosing started at 8th day after the surgery.The treatment duration were divided into 2groups;14 days and 28 days of dosing,the sham operation and the model groups were given normal saline with a standard of 0.72 ml/100 g body weight per day.The esomeprazole group was dosing with esomeprazole daily at a dose of 0.36 ml/100 g body weight.Rats in Shuganlidanhewei group were given Shuganlidanhewei oral with daily dosage 0.72 ml/100g body weight.At the end of the treatment of two groups,the rats were sacrificed and the samples were taken.In order to compare the ability of the Shuganlidanhewei in treating reflux esophagitis with ezomeprasole and the model group by analyzing the results of the measurement of the lower esophagel pH value.Moreover,compare the histological changing of esophageal mucosa in each group(it was observed through microscope investigation method).In addition,using enzyme-linked immunosorbent assay(ELISA)to detect IL-8 in esophageal tissue of rats with reflux esophagitis.Results:1.Lower esophageal pH value in the model group is lower than the sham operation group’s(14 days:2.48+0.66;28 days:2.33+0.83).Compared with the model group, Shuganlidanhewei group can increase lower esophageal pH value(P<0.05),(14 days:4.65+1.55;28 days:5.19+1.84);Shuganlidanhewei group and esomeprazole group increase the lower esophageal pH There was no significant difference in values(P>0.05).2.Compared with the sham operation group,the esophaeal mucosa of rat in model group has pale,coarse,damaged mucosa membrane,squamous epithelium cell hyperplasia,elongation of the lamina propria papilae;Compare with the model group,the esophageal mucosa appearance of animal model in treatment group was obviously ameliorated.The ameliorated result of treatment period 28 days in treatment group was obviously better than the 14 days treatment period.The result of ameliorated esophageal tissue at the same treatment period was shown no significant different between Shuganlidanhewei and esomeprazole groups.3.Compared with the sham operation group,the IL-8 values in the esophageal tissue of the model group(14 days:47.39+5.66;28 days:48.43+6.04)was significantly increased(P<0.05).The levels of IL-8 in each treatment group was decreased(P<0.05),and the IL-8 values decreased significantly after 28 days of treatment(P<0.05)by using the model group as a mean.The IL-8 values(pg/ml)between Shuganlidanhewei decoction group(14 days:40.75+4.20;28 days:36.70+2.87)and esomeprazole group(14 days:40.16+1.83;28 days:36.27+3.97)were compared.As a result,there was no significant difference(P>0.05).Conclusion:1.Shuganlidanhewei decoction can regenerate the esophagus tissue which is damaged from reflux esophagitis in vivo by increasing lower esophageal pH value.The ameliorated esophageal mucosa from using Shuganlidanhewei has the same result as esomeprazole’s treatment.We conclude that Shuganludanhewei has a treatment efficiency with reflux esophagitis.2.Shuganlidanhewei decoction has decreasing IL-8 values in esophageal mucosa.It is possible that the mechanism in treating reflux esophagus of Shuganlidanhewei may be related to the decreasing of IL-8 values in esophageal mucosa. |
