Purpose: To observe the levels of various lipid profiles of Qingzhi Tongmai Granules in experimental atherosclerosis model rats,and compare the ratios of drugs with the best curative effect.Furthermore,the relationship between the expression of PPARγ,LXRα and CD36 in the heart and liver and the level of lipid metabolism was studied by Qingzhi Tongmai Granule,and the mechanism of its treatment of atherosclerosis was discussed.Material and method:1.The experimental animals selected in this experiment were144 male Sprague-Dawley rats.During the experiment,the animals were grouped by random number table.144 rats were randomly divided into 12 groups(normal group,model group,Chinese medicine group 10 groups),12 in each group.After each group of rats under the same experimental conditions,after one week of adaptive feeding,the rats in the normal group were subjected to AS modeling.The model was established by intraperitoneal injection of vitamin D3(600,000 IU/kg)and compound high-fat diet.Feeding,while the normal group of rats were injected intraperitoneally with the same amount of normal saline and normal feed.2.After the end of the experiment at the 9th week,the abdominal aorta blood was taken after fasting for 12 hours,and the serum cholesterol(TC),triglyceride(TG),low density lipoprotein(LDL-C)and the serum of each group were detected.High density lipoprotein(HDL-C)content.After the rats were completely anesthetized,the liver and heart tissue were frozen for use.3.The statistical data of 12 groups of experimental data were analyzed by SPSS17.0 software.The measurement data of each group were analyzed by one-way ANOVA.The data were expressed as mean±standard deviation(x±s).The results were not statistically significant.P <0.05 was considered statistically significant,and P < 0.01 was considered significant.4.Real-time fluorescence quantitative PCR and Western blot were used to detect the expression levels of PPARγ,LXRα and CD36 in the heart and liver tissues of rats(normal group,model group and optimal dose group).analysis.At the same time,the levels of apoA1 and ox-LDL in rat plasma were detected by enzyme-linked immunosorbent assay(ELISA)and the experimental data were statistically analyzed.Results:1.The results of four blood lipid tests showed that compared with the normal group,the contents of TC,TG and LDL-C in the model group were significantly increased(P<0.01),and the HDL-C content was significantly decreased(P<0.01).Compared with the model group,the TC content of the Chinese medicine group was significantly lower in the 3,7,8,9,and 12groups(P<0.05),and the TG content in the Chinese medicine group except the fifth group was significantly lower(P<0.05).The levels of HDL-C in groups 6,8,11 and 12 were significantly increased(P<0.05),and the contents of LDL-C in groups 3,7,8,10 and 12 were significantly lower(P<0.05).The remaining groups were TC and TG.LDL-C content decreased slightly and HDL-C increased slightly,but it was not statistically significant(P>0.05).Therefore,after comprehensive comparison,it was found that the 8 and 12 groups were statistically significant among the four indicators,and were selected as the optimal dose group for further experiments.2.The content of apoAI and ox-LDL in plasma was detected by ELISA.According to the statistical analysis,the plasma apoAI content in the model group was significantly lower than that in the normal group(P<0.01).Compared with the model group,the Chinese medicine group 8 and group 12 The plasma apoAI content was significantly increased(P<0.01).Compared with the normal group,the plasma ox-LDL content in the model group increased significantly(P<0.01).Compared with the model group,the plasma ox-LDL levels in the Chinese medicine group 8 and group 12 were significantly lower(P<0.05).3.The results of Real-time fluorescence quantitative PCR showed that the expression of PPARγ and LXRα in heart and liver tissues was significantly lower than that in the normal group(P<0.05).The content of CD36 in heart and liver tissues was significantly increased.(P<0.05).Compared with the model group,the expression of PPARγ and LXRα in the heart and liver tissues of the Chinese medicine group(8 groups and 12 groups)was significantly increased(P<0.05).Compared with the model group,the mRNA expression of CD36 in the liver tissue of the Chinese medicine group was compared.Significantly decreased(P<0.01),the expression of CD36 mRNA in heart tissue decreased,but the 8 groups did not have statistical significance(P>0.05),the 12 th group had statistical significance(P<0.05).4.Western-blot results showed that compared with the normal group,the expression of PPARγ,LXRα,protein in the heart and liver decreased significantly in the model group(P<0.05);the content of CD36 in the heart and liver tissues increased significantly(P<0.05).<0.05).The protein expression of PPARγ in heart and liver was increased by model group and Chinese medicine group(8 groups and 12 groups)(P<0.05);protein expression of LXRα in liver tissue was significantly increased(P<0.01),LXRα was Although there were improvement in heart tissue,8 groups were statistically significant(P<0.01)and 12 groups were not statistically significant(P>0.05).Compared with the model group,the protein of CD36 in heart and liver tissues was compared with the model group.The expression decreased(P<0.01).Conclusion:1.After the blood lipid test indicators confirmed,the lipid-lowering effect of Group 8 and Group 12 was relatively better than other Chinese medicine groups.2.Through classical modeling(high-fat diet-fed rats and vitamin D3 intraperitoneal injection),it was found that TC,TG,LDL-C and ox-LDL were significantly elevated in the model group,and HDL-C was significantly decreased.This indicates that the rat model of hyperlipidemia arteriosclerosis was successfully modeled.3.Qingzhi Tongmai Granule can increase the content of apoAI in plasma of atherosclerotic model rats;the content of ox-LDL in plasma decreases.4.Qingzhi Tongmai Granule can increase the expression of PPARγ and LXRα mRNA and protein in heart and liver tissues of atherosclerotic model rats;the expression of CD36 mRNA and protein is decreased.5.The regulation of cholesterol transport by Qingzhi Tongmai Granule is due to the increase of mRNA and protein expression of PPARγ and LXRα.6.Qingzhi Tongmai Granule can reduce the expression of CD36 mRNA and protein,reduce the endocytosis of oxidized lipid and reduce the formation of foam cells.7.The mechanism of Qingzhi Tongmai Granule in preventing and treating atherosclerosis may be through increasing the expression of PPARgamma and LXRalpha in heart and liver tissues,increasing the content of apoAI in blood,promoting the level of HDL,promoting the transport of free cholesterol,inhibiting the absorption of cholesterol and accelerating themetabolism of cholesterol,and by reducing the expression of CD36 gene and protein.Decreasing the content of ox-LDL in vivo and decreasing lipid endocytosis can reduce foam cell production and promote lipid outflow.Thus,the formation and development of atherosclerosis can be delayed. |
