Experimental Study On Effects Of Qingzhi Tongmai Granule On Blood Lipid And VEGF And IFN - γ In Rats With Atherosclerosis

Purpose:Observation the influence of Qingzhi Tongmai Granule on experimental hyperlipemia model rats blood fats level and the expression of VEGF and IFN-γ.Explore on the mechanism of Qingzhi Tongmai Granule in preventing and treating atherosclerosis(AS),Furtherly providing theoretical basis for AS resistance on TCM.Material and method:1.Select 48 healthy and clean male Wistar rats,divided into 6 groups randomly with 8 in each, namely(1)normal group;(2)model group;(3)the western medicine control group;(4)high-dose of Qingzhi Tongmai Granule group;(5)middle-dose of Qingzhi Tongmai Granule group;(6) low-dose of Qingzhi Tongmai Granule group.In addition to the normal group,the others were injected Vit D3 with high fat feed.2.At 16 th week the end of experiment,detected the content of blood serum of TC,TG,LDL-C,HDL-C in the rats model,measured the expression levels of VEGF and determination of IFN-γ by the method of enzyme-inked immunosorbent(ELISA).Results:1.After injecting Vit D3 one-off,with high fat feed compounded,the content of TC,TG,LDL-C were significantly increased,and HDL-C decreased significantly,the result show that hyperlipidemia rat model is established successfully.2.Compared with normal group, content of TC,TG,LDL-C increased,HDL-C decreased,both significantly in model group(P<0.01). Compared with model group, simvastatin group, highdose and middle-dose of Qingzhi Tongmai Granule could bring TC,TG,LDL-C down obviously(P<0.01).There is no significant difference found between simvastatin group and middledose of Qingzhi Tongmai Granule at level of TC,TG,LDL-C(P>0.05).However, each dose group of Qingzhi Tongmai Granule had no obvious effect on the expression of HDL-C.3.ELISA results showed that: Compared with normal group,expression level of VEGF and IFN-γ in model group had an incredible increase(P<0.01).Compared with model group, Simvastatin group, high-dose and middle-dose of Qingzhi Tongmai Granule could reduce content of VEGF, IFN-γ in serum, with significant difference(P<0.01).Simvastatin group and middle-dose of Qingzhi Tongmai Granule showed similar efficacy on expression level of VEGF, IFN-γ(P>0.05).Conclusion:1.Qingzhi Tongmai Granule could decrease the content of TC,TG,LDL-C,having no significant difference from Simvastatin group.2.QingzhiTongmai Granule could inhibited the expression level of VEGF and IFN-γ significantly.3.The effect on Qingzhi Tongmai Granule about prevention and treatment of atherosclerosis, maybe through regulating blood lipid metabolism,reducing the levels of VEGF and IFN-γ, Inhibiting the inflammation for AS,protecting vascular endothelium.
