Purpose:To explore the effect of acupuncture on the balance mechanism of Th1/Th2 in type I hypersensitivity and type IV hypersensitivity.Material and method:110 SPF-grade 8-week-old male SD rats were randomly divided into 10 normal skin sensitized rats(NG),10 ear sensitized rats(RM),10 positive ear sensitized rats(RY),10 ear sensitized rats(RC)and 10 foot sensitized rats according to a computer-generated random number table after one week of adaptive feeding.10 rats in the ZM group,10 rats in the ZY group,10 rats in the ZC group,10 rats in the PM group,10 rats in the PY group and 10 rats in the PC group.Another 10 mice were fed normally to prepare anti-ovalbumin serum.All rats were fed at room temperature,fed routinely and steamed twice.10 rats in RY group,ZY group and PY group were given loratadine 0.2mg dissolved in 1 ml of 10.9% sodium chloride for intragastric administration,RC group,ZC group and PC group were given nanoneedles(0.25 mm x 13mm)to Quchi point of rats.Xuehai acupoint,retain needle for 30 minutes,during the period of acupuncture,no reinforcing and reducing manipulation,no electricity,once a day,continuous intervention for 7 days.After 7 days,auricular swelling model was replicated in the control group(RM),the positive drug group(RY)and the acupuncture group(RC).The foot sensitization model was replicated in the control group,the foot sensitization model was positive in the drug group,and the foot sensitization model was replicated in the acupuncture group.The skin sensitization model was positive in the skin sensitization reaction model control group and the skin sensitization model was positive in the skin sensitization reaction model group.Drug group and skin sensitization model group were used to replicate the skin sensitization model.After the completion of the model,artery blood was taken from rats.Serum IgE level was measured by Elisa method.The locus coeruleus of rat skin was taken out by perforator.The inhibition rate of locus coeruleus was calculated by spectrophotometry.The auricle of rat was taken out by perforator,and the swelling degree of auricle was calculated by infiltration method.Immunohistochemical method was used to detect the expression of IL-2,IL-4,IL-10 and IFN-gamma in skin,auricleand foot of rats.Results:1.Compared with the skin sensitization model group,the serum IgE level in the skin sensitization model group was lower in the skin sensitization model group than in the skin sensitization model group(P<0.05).Compared with the skin sensitization model positive drug group,the serum IgE level in the skin sensitization model group was lower(P<0.05).There was no difference in the serum IgE level between the auricle sensitization model and the foot sensitization model(P>0.05).2.Compared with the control group of auricle sensitization model,the auricle swelling degree of the positive drug group of auricle sensitization reaction model and the acupuncture group of auricle sensitization reaction model were lower and the inhibition rate of auricle swelling degree was higher(P<0.05).Compared with the positive drug group of auricle sensitization reaction model,the auricle swelling degree of the acupuncture group of auricle sensitization reaction model was lower and the inhibition rate of auricle swelling degree was higher(P<0.05).3.Compared with the foot sensitization model control group,the foot swelling degree in the foot sensitization model positive drug group and the foot sensitization model acupuncture group was lower(P<0.05).Compared with the foot sensitization model positive drug group,the foot swelling degree in the foot sensitization model acupuncture group was lower(P<0.05).4.Compared with the normal control group(left),the levels of IL-4 and IL-10 decreased and the levels of IL-2 and IFN-gamma increased in the ear sensitization model group(ringht),the ear sensitization model positive drug group and the ear sensitization model acupuncture group(P<0.05).Compared with the ear sensitization model control group,the levels of IL-4 and IL-10,IL-2 and IFN-gamma increased in the ear sensitization model positive drug group and the ear sensitization model acupuncture group,and the levels of IL-2 and IFN-gamma increased.The levels of IL-4 and IL-10 were higher in the EAR model group,while the levelsof IL-2 and IFN-gamma were lower in the EAR model group than those in the EAR model positive group(P<0.05).5.Compared with the control group(left),the levels of IL-4 and IL-10 in the foot sensitization model group(right),the foot sensitization model positive drug group and the foot sensitization model acupuncture group decreased,while the levels of IL-2 and IFN-gamma increased(P<0.05).Compared with the foot sensitization model control group,the foot sensitization model positive drug group,the foot sensitization model acupuncture group IL-4 and IL-10 water.The levels of IL-4 and IL-10 in the foot sensitization model group were higher than those in the foot sensitization model group,while the levels of IL-2and IFN-gamma were lower than those in the foot sensitization model group(P<0.05).6.Compared with the normal control group of skin sensitization model,the levels of IL-4 and IL-10 in the skin sensitization model group,the skin sensitization model positive drug group,the skin sensitization model acupuncture group increased,the levels of IL-2 and IFN-gamma decreased(P<0.05).Compared with the skin sensitization model control group,the skin sensitization model positive drug group and the skin sensitization model acupuncture group IL-4.The levels of IL-4 and IL-10 decreased,while the levels of IL-2 and IFN-gamma increased(P<0.05).Compared with the skin sensitization model positive drug group,the levels of IL-4 and IL-10 in the skin sensitization model acupuncture group were lower,while the levels of IL-2 and IFN-gamma were higher(P<0.05).7.Compared with the normal control group of skin sensitization model,the level of blue spot light density in the skin sensitization model group,the skin sensitization model positive drug group and the skin sensitization model acupuncture group increased,and the inhibition rate of blue spot was lower(P<0.05).Compared with the skin sensitization model control group,the skin sensitization model positive drug group and the skin sensitization model acupuncture group were better than the skin sensitization model control group.The blue spot light density water was lower and the inhibition rate of blue spot was higher(P<0.05).Compared with the positive drug group of skin sensitization reaction model,the blue spot light density water of skin sensitization reaction model acupuncture group was lower and the inhibition rate of bluespot was higher(P<0.05).Conclusion:1.Skin allergic reaction belongs to type I hypersensitivity,auricle and foot allergic reaction belongs to type IV hypersensitivity.Acupuncture has obvious improvement effect.2.Acupuncture can reduce the secretion of Th 2 in type I hypersensitivity reaction,reduce the secretion of Th1 in type IV hypersensitivity reaction,maintain the balance of Th1/Th 2,and have obvious bidirectional regulation.
