Purpose:To clarify the effect of Shenxian Shengmai Oral Liquid on sinus node fibrosis in aging rats and to explore the effect of TGF-?1/Smad pathway in improving the sinus node fibrosis in aging rats.mechanism.Material and method:There were 60 SPF grade SD rats,including 20 3 months old rats,20 12 months old rats and 20 18 months old rats.They were randomly divided into 2 groups: Shenxian Shengmai Oral Liquid intervention group(experimental group).)and normal controls(controls).Rats in the control group were fed conventionally;rats in the experimental group were regularly given intragastrically Shenxi Shengmai Oral Liquid 2 ml.The heart rate of each group of rats was measured at the beginning of the experiment,on the 7th day,the 14 th day,and the 21 st day,and the material was taken after 30 days.HE staining was used to observe the histopathological changes of sinoatrial node;Masson staining was used to observe sinus node fibrosis;Western-blot was used to detect the expression of age-related gene Klotho;Western-blot and q PCR were used to detect TGF-?1,Smad 2,Smad 3,and Smad.7 Protein and m RNA expression levels.Results:1.Heart rate: In the control group,the heart rate decreased significantly in the 12-month-old group and the 18-month old group,and the sinus node tissue fibrosis was severer.The heart rate and sinus node fibrosis did not change significantly in the 3-month-old group.In the experimental group,The heart rate of the 12-month-old group and the 18-month-old group increased significantly compared with the initial heart rate(P<0.05),and the degree of sinus node tissue fibrosis decreased compared with the age-matched control group.2.Histology: In the control group,the pathological changes and fibrosis of the sino-atrial node were severe in the 12-month-old and 18-month-old rats.The pathological changes and fibrosis of the sino-atrial node were not significantly changed in the 3-month-old group;The 12-month old group and the 18-month old group were compared with the control group.The pathological changes and fibrosis of the sinus node were reduced.3.Klotho: In each group of rats,Klotho protein levels were high in the sino-atrial node of 3-month-old rats,and Klotho protein expression levels in the sinus node of rats were significantly lower in the 12-month-old and 18-month-old groups than in the 3-month old group.(P<0.05);Compared with the control group,the expression of Klotho protein in the sinus node of the experimental group decreased less.4.TGF-?1/Smad pathway: In the control group,the expression levels of TGF-?1,Smad 2 and Smad 3 increased with the age,and the Smad 7 expression level decreased(P<0.05);the experimental group compared with the control group,in December The expression levels of TGF-?1,Smad 2 and Smad 3 were significantly decreased and the expression of Smad 7 was increased in the age group and the 18-month-old group(P<0.05),while there was no significant difference between the experimental group and the control group in the 3-month-old group(P> 0.05).Conclusion: 1.With the increase of age,the number of sinus node tissue fibrosis increases and the rate of heart rate decrease increases.2.Shenxian Shengmai Oral Liquid can increase heart rate and improve sinus node tissue fibrosis.3.The TGF-?1/Smad pathway plays an important role in improving the sinoatrial node tissue fibrosis in Shenxian Shengmai Oral Liquid and delays the progression of sinus node fibrosis.
