Purpose: To observe the change of the expression of calcium ion channel in the improvement of the dysfunction of the sinus node in the elderly rats,To explore the mechanism of Shenxian shengmai oral liquid to improve the function of sinus node in old rats.Material and method:After the rat heart rate and ECG monitoring were screened,the rats were divided into three groups: the blank control group(control group),the sick sinus syndrome group(sinus syndrome group),the shenxian shengmai oral liquid treatment group(shenxian shengmai treatment group).In the control group,rats with normal heart rate were given 2ml/d.The group of sinus atrial node syndrome was the rats with pathological sinus syndrome,and the normal saline was 2ml/d.In the treatment group,the heart rate decreased with the rats with the pathological sinus syndrome,and the oral liquid was administered in 2ml/d for 30 days.In the 7th,14 th and 21 st days,MD3000 biological signal collection and processing system was used to record the heart rate changes of rats.After 30 days,HE staining was used to observe the pathological changes of rat sinus node in rats.Western blot was used to determine the expression of Cav1.2,Cav1.3 and Cav3.1,Cav3.2 and Cav3.3 protein in the calcium ion channels of rat sinus node in rats.Results: 1.MD3000 biological signal acquisition and processing system recorded the heart rate of rats,and compared with the control group,the heart rate of the rats in the sinus node syndrome group decreased significantly.Compared with the sinus node syndrome group,the heart rate of the rats in shenxiansheng group increased significantly(P<0.05).2.HE staining showed that,compared with the control group,cellular swelling,intercellular fissures widen,nuclear hyperchromatism,the pathological changes of the sinus node in the sinus node were more serious.Compared with the sinus node syndrome group,liver celhterl swelling,intercellular fissures have been restored,the nucleus becomes lighter,the pathological changes of the sinus node in the treatment group were significantly improved.3.Western blot showed that,compared with the control group,Cav1.2,Cav1.3 and Cav3.1,Cav3.1 and Cav3.1 were significantly reduced in the rat sinus node of rat sinus node.In comparison with the sinus node syndrome group,Cav1.2,Cav1.3 and Cav3.1,Cav3.1,Cav3.1 and Cav3.3 protein expression were significantly increased in the rat sinus node(P<0.05).Conclusion: 1.The heart rate of the elderly rats with the pathological sinus syndrome was significantly increased by the Shenxian shengmai oral liquid.2.Shenxian shengmai oral solution significantly improved the pathological changes of the sinus node in the elderly rats with pathological sinus syndrome,thereby improving the dysfunction of the elderly rat sinus node.3.The expression of Cav1.2,Cav1.3 and Cav3.1,Cav3.2 and Cav3.3 in the calcium channels in the rat sinus node of the elderly rats were improved by the Shenxian shengmai oral liquid,to improve the self-regulation of sinus node cells,thus improving the dysfunction of the sinus node in elderly rats.
