Objective:Schisandra chinensis?Turcz.?Baill,the dried ripe fruit of Schisandra chinensis?Turcz.?Baill.According to traditional Chinese medicine,it tastes sweet and sour,warm in nature,enters into lung,heart,and kidney meridians,it has the effect of astringency,it benefits vital energy,promotes the production of body fluids,nourishes the kidney as well as calms the mind.Clinically,it is mainly used to treat chronic cough,asthma,wet dream,urinary frequency,diarrhea,sweating,thirst,heart palpitation,insomnia,etc[1].The most common processed products that are raw Schisandra chinensis fruits,wine Schisandra chinensis fruits and vinegar Schisandra chinensis fruits.According to the“Traditional Chinese medicine theory”,raw Schisandra chinensis fruits is used to treat coughs and the processed products are used for compensation[2],meaning of raw and processed products have different medical effects.This paper is from the aspect of change of 5-HMF content of different steaming time of Schisandra chinensis fruits before and after processing and different processed products,comparing different components of the volatile oil in processed products of Schisandra chinensis fruits,studying on the pharmacological effects of different processed products of Schisandra chinensis fruits,so as to provide pharmacological basis and experimental reference for the clinical reasonable application for the“processed Schisandra chinensis fruits as a tonic”.On the basis of the previous research group,this paper studies the comparison of the chemical composition of different processed products of Schisandra chinensis and the effect of"into the tonic"in order to provide experimental reference for the rational use of“processed products are used for compensation”.Materials and methods:Materials:Schisandra chinensis fruits,purchased from Da Lishu GAP base,Dandong,Liaoning province,identified by Professor Wang Bing from medicinal plants teaching and research section of pharmaceutical college of Liaoning University of Traditional Chinese Medicine,is the dried ripe fruit of Schisandra chinensis?Turcz.?Baill.Methods:1.Changes of the content of five hydroxymethyl furfural?5-HMF?before and after Schisandra chinensis fruits processed products.High performance liquid chromatography?HPLC?was used to determine the content of5-HMF?5-HMF?in raw Schisandra chinensis fruits,wine Schisandra chinensis fruits and vinegar Schisandra chinensis fruits.2.Change of 5-HMF content of different steaming time of Schisandra chinensis fruits different processed products.The content of five hydroxymethylfurfural?5-HMF?in different processed products of Schisandra chinensis fruits was determined by HPLC with five hydroxymethylfurfural?5-HMF?content as index and different steaming time as the investigation factors.3.Analysis of volatile oil in different processed products of Schisandra chinensis fruits by GC-MS.The components of volatile oil from Schisandra chinensis fruits,wine Schisandra chinensis fruits and vinegar Schisandra chinensis fruits were tested by GC-MS,and the total ion flow chart was obtained.The mass spectra of the peaks in the total ion flow chart were scanned by mass spectrometry.4.Study on the effect of different kinds of processing products of Schisandra chinensis fruits on yang kidney deficiency model rats.The model of yang kidney deficiency rats were induced by methimazole system,and were randomly divided into model group,Yougui Pill group,raw Schisandra chinensis fruits group,wine Schisandra chinensis fruits group,vinegar Schisandra chinensis fruits group,as well as blank control group with 10 rats in each group.After 30 days'intragastric administration,put to death in the rat,to measure serum FT3,FT4,TSH,NO;to measure plasma cAMP and cGMP content and calculate the ratio of cAMP/cGMP;to observe the thyroid pathological changes in rats.5.Study on the therapeutic effect of different processed products of Schisandra chinensis fruits on rat model of kidney deficiency deficiency.The rat model of kidney essence deficiency were induced by Tripterygium Glycosides tablet,and were randomly divided into model group,raw Schisandra chinensis fruits group,wine Schisandra chinensis fruits group,vinegar Schisandra chinensis fruits group,as well as blank control group,a total of 5 groups,each group of 10.After 30 days'intragastric administration,rats were killed,then measured the quality of the epididymis and renal tissue,the content of serum INH-B?FSH in rats was determined by ELISA;observed the spermatogenic cells pathological changes in rats?the pathological morphology of rat testis was observed under the microscopecal,culated the spermatogenic cells in the Johnson score and the apoptosis rate of spermatogenic cells?AR?.,and the compared therapeutic effect of different kinds of processing products of Schisandra chinensis fruits on kidney essence deficiency model rats.6.Study on the effect of different processed products of Schisandra chinensis fruits on the repair of damaged renal cells induced by hydrogen peroxide.Selected renal cells model to determine the proliferation rate of different processed products of Fructus Schisandra were screened by CCK-8 method,screen out the optimal dosing concentration.Renal cell were induced by hydrogen peroxide?H2O2?,After the damage to the kidney cells is successful,given the best concentration of Schisandra chinensis fruits in different artillery products,ELISA kits were used to determine the cell supernatant of bone morphogenetic protein 7?BMP-7?,hepatocyte growth factor?HGF?and vascular endothelial growth factor?VEGF?content,in order to compare the repair effect of different processed products of Schisandra chinensis on damaged renal cells.7.Study on the effects of different processed products of Schisandra chinensis fruits on LPS stimulated macrophages in RAW264.7 mice.The RAW264.7 mouse macrophage was selected as the cell model in vitro,and the model of lipopolysaccharide?LPS?was used to build the model.The CCK-8 method was used to determine the proliferation rate of RAW264.7 mice stimulated by different products of Schisandra chinensis fruits by LPS,and the best dosage of Schisandra chinensis fruits was screened.The relative phagocytosis rate of different processed products of Schisandra chinensis with different concentrations on LPS stimulated RAW264.7 mouse macrophages was measured by the enzyme labelled instrument.The contents of TNF-,IL-I,and NF-kB in supernatants of macrophages stimulated by LPS were determined by ELISA,and the effects of different processed products of Schisandra chinensis fruits on LPS stimulated RAW264.7mice macrophages were determined by RAW264.7 assay.Results:1.Changes of the content of five hydroxymethyl furfural?5-HMF?before and after Schisandra chinensis fruits processed products.Schisandra chinensis fruits can produce Maillard reaction during processing.It can produce a new component,five hydroxymethylfurfural?5-HMF?.2.Change of 5-HMF content of different steaming time of Schisandra chinensis fruits different processed products.Steamed Schisandra chinensis fruits with different steaming time,and different processed products of Schisandra chinensis fruits were obtained.The content of five hydroxymethyl furfural?5-HMF?in Schisandra chinensis fruits was different.With the prolongation of the steaming time,the content of five hydroxymethyl furfural?5-HMF?in wine Schisandra chinensis fruits and vinegar Schisandra chinensis fruits is more and more.3.Analysis of volatile oil in different processed products of Schisandra chinensis fruits by GC-MS.The content of volatile oil in different processed products of Schisandra chinensis fruits is different.After processing,the content of volatile oil is reduced,and the components of volatile oil in different processed products of Schisandra chinensis fruits are different.4.Study on the effect of different kinds of processing products of Schisandra chinensis fruits on yang kidney deficiency model rats.Compared with the model group,the Schisandra chinensis fruits processed products are from different extent increased in rat serum FT3,FT4,NO and TRH in the hypothalamus,decrease the serum level of TSH rats?P<0.05,0.01?;increased the content of cAMP in rat plasma,decreased the plasma level of cGMP?P<0.05,0.01?,so the ratio of cAMP/cGMP increased;between the different processed products of Schisandra chinensis fruits,compared with the optimal effect of wine Schisandra chinensis fruits?P<0.01?;improved the effect of the processed products of Schisandra chinensis fruits on yang kidney deficiency model rats were thyroid pathological changes,and the effect of wine and vinegar Schisandra chinensis fruits was better than that of the raw Schisandra chinensis group.5.Study on the therapeutic effect of different processed products of Schisandra chinensis fruits on rat model of kidney deficiency deficiency.Compared with the model group,the quality of epididymis and renal of rats in each group were increased after administration of?P<0.01?;compared with the model group,the treatment group all can enhance the different levels of serum INH-B and serum FSH level?P<0.01?,the effect of wine Schisandra chinensis fruits is the best;the pathological morphology ofdifferent Schisandra chinensis fruits processed products of all can improve the deficiency of the kidney essence deficiency model of rat testis,wine Schisandra chinensis fruits and vinegar Schisandra chinensis fruits is better than raw Schisandra group.6.Study on the effect of different processed products of Schisandra chinensis fruits on the repair of damaged renal cells induced by hydrogen peroxide.With the increase of drug concentration,the effect of different processed products of Schisandra chinensis fruits on the rate of renal cell proliferation was increasing.The highest rate of renal cell proliferation in 500?g/mL-1,and you can see the crude rate of group was higher than that of the product proliferation,the highest proliferation rate of proliferation wine Schisandra chinensis fruits;compared with the model group,each group of BMP-7,VEGF and HGF were significantly increased;compared with the crude products between the groups the content of HGF,BMP-7,VEGF were increased significantly.7.Study on the effects of different processed products of Schisandra chinensis fruits on LPS stimulated macrophages in RAW264.7 mice.The promotion of the proliferation rate of macrophages in RAW264.7 mice showed an upward trend.When the concentration was 100?g/mL-1,the proliferation rate of macrophages in RAW264.7 mice was the highest.It can be seen that the proliferation rate of the artillery products is higher than that of the various raw materials,and the multiplication rate of wine Schisandra chinensis fruits is the highest.With the increase of drug concentration,the phagocytosis promoting effect of different processed products of Schisandra chinensis fruits on the macrophages of RAW264.7 mice showed a decreasing trend.When the concentration was 500 ug/mL-1,the phagocytosis rate of macrophages in RAW264.7 mice was the highest,and it could be seen that the phagocytosis rate of processed products was higher than that of all the raw products group,and the rate of phagocytosis of wine Schisandra chinensis fruits was the highest.The TNF-,IL-1 and NF-kB levels in the model group decreased significantly.Compared with the model group,the levels of TNF-alpha,IL-1 beta and NF-kB increased significantly compared with the model group.Compared with the raw group,the contents of TNF-,IL-1,beta and NF-kB increased significantly in the processed products group.Conclusion:After steaming?wine steaming or vinegar steaming?,the new ingredient 5-HMF was added,and the maillard reaction was speculated in the process of Schisandra chinensis fruits.With the increase of evaporation time,the content of 5-HMF is increased,which can be used as the judging index of processing degree.After processing,the volatile oil content of Schisandra chinensis fruits decreased and the composition of volatile oil changed.The results showed that the wine Schisandra chinensis fruits was better than raw Schisandra chinensis fruits and vinegar Schisandra chinensis fruits with obvious beneficial effect,so“processed Schisandra chinensis fruits as a tonic”should use wineSchisandra chinensis fruits.
