Purpose:To observe the clinical effect of YuKui formula in active ulcerative colitis patients with spleen-kidney yang deficiency and damp-heat in their intestine,and by analyzing the expression of claudin-1 and STAT3 in intestinal mucosa before and after taking YuKui formula,to investigate its therapeutic mechanisms.Material and method:Collecting the patients from the department of traditional Chinese medicine in Shenyang Military District General Hospital during from June 2016 to June 2017.Select 40 patents that meet the diagnostic criteria,inclusion criteria and exclusion criteria.The 40 patients were randomly divided into two groups,and there were 20 cases in the treatment group and 20 cases in the control group.The treatment group was given YuKui formula,however,the control group was given Mesalazine.The course of treatment was 8weeks.Adopting the TCM syndrome quantification table and intestinal mucosal grading table to score.all the patients were performed electronic colonoscopy before and after treatment,and in treatment group selecting 10 patients randomly and taking mucosal tissue in obvious lesion to detect the expression of claudin-1 and STAT3 protein in immunohistochemistry.Recorded all the side effects of two groups during the treatment and measured liver function of patients in treatment group after treatment.SPSS17.0 statistical software was used to analyze the data.The count data were measured by X~2 test.The measurement data were measured by T test and the nonparametric rank sum test was used for the grade data.Results:1.Efficacy evaluation1.1 TCM syndrome efficacy evaluation:The two groups patients'main symptoms were improved obviously after treatment,and the difference between the two groups was not statistically significant(P>0.05).The treatment group was significantly better than the control group on secondary symptoms(P<0.05).The total curative effect and total effective rate of TCM syndrome in treatment group were better than that in control group(P<0.05).1.2 Efficacy evaluation of electronic colonoscopy:after treatment,the mucosa of the two groups was significantly improved,and the difference between the two groups was not statistically significant(P>0.05)2.Safety evaluationNo obvious adverse reactions occurred in the two groups during the treatment,and the liver function was normal in treatment group after treatment.3.After treatment,the STAT3 protein was down-regulated and claudin-1 protein was up-regulated in the treatment group,the difference was statistically significant(P<0.05).Conclusion:1.YuKui formula was effective and safe,and it was worthy of clinical application.2.The therapeutic mechanisms of YuKui formula may be related to inhibit the activation of signaling pathway and improve the damaged intestinal mucosal barrier.
