Purpose:To observe the efficacy and safety of ribonucleic acid ? in combination with injection of cindividin into the abdominal cavity for the treatment of malignant pleural effusions.The two drugs were tested for susceptibility to malignant pleural effusion and ascites fluid extraction in vitro,and preliminarily explored the predictors of efficacy.Material and method:MTT assay for the inhibition of growth of breast cancer cells(MCF-7,MDA-MB-435,MDA-MB-231)by RNA ? and RNA ? combined with cinobufacin at different concentrations,and collecting cell morphology data,Compare the cell morphology changes after different concentrations of drugs.Seventeen patients with malignant pleural effusion or ascites were randomly individe into two groups: Ribonucleic acid combined with Cinobufacini and Cinobufacini monotherapy.Paracentesis-free interval(PaFI)were detected,side effects records and chemosensitivity test results were collected.Patients with malignant pleural effusion and ascites fluid were collected for selection of tumor cells before and after treatment.The effects of RNA ? on the proliferation and metastasis of tumor cells were collected.Immune parameters(CD3,CD4,CD4/CD8),and blood before and after treatment of patients with malignant pleural effusion were collected.Routine(white blood cells,neutrophils,platelets),liver function,renal function,blood lipids,blood glucose and other indicators.Results: 1.The combination of RNA ? and cinobufacini can inhibit the growth of breast cancer cells and is more cytotoxic than ribonucleic acid ?.2.The morphological changes of RNA ?,RNA ? combined with Huachanin include rounding,shrinking,deformation and floating.3.All the 17 patients with malignant thoracic and abdominal cavity water had symptomatic remissions.The tumor inhibition rates of ribonucleic acid ? combined with cindanin and monocerebroins were 49.8% and 39.8%,respectively.The combination group was significantly higher than the single drug group(P).<0.05).The median follow-up timewas 32 weeks.4.The proportion of patients with Pafi ? 8 weeks of Huachansu monotherapy group and ribonucleic acid ? combined Huachansu group was 25% and 66.7%,and the median PaFI was 12 weeks in the combined group.In the group of 7 weeks,the combination group had a prolonged trend(P<0.05)compared with the monotherapy group5.In adverse reactions,the incidence of adverse reactions such as abdominal pain associated with drugs was 11.1%,which was lower than that of the single drug group,which was 62.5%;Compared with PaFI <8 weeks,the combined effect of the two drugs in patients with long-term effective PaFI ? 8 weeks is higherConclusion:1.Intravenous administration of ribonucleic acid ? in combination with cinobufacrine has a synergistic effect in inhibiting systemic tumors and reducing malignant pleural effusions;2.The ribonucleic acid ? combined group has a certain attenuating effect during the course of treatment;3.In vitro drug susceptibility inhibition was high in the combination group,suggesting and demonstrating a good prognosis in the combined group chest cavity administration.
