Objective Colon cancer is the leading cause of death worldwide,representing a major threat to public health.To investigate the expression of Oct4 protein in colon cancer and analyze the prognostic value of Oct4 in colon cancer.Methods From January 2008 to December 2011,89 specimens of left hemicolectomy were selected and 77 specimens of right hemicolectomy were selected.There were 70 males and 96 females,with a male to female ratio of 0.73 to 1 and an average age of63.7 ± 14.1 years old.All the patients did not receive any radiotherapy or chemotherapy before surgery,and the postoperative pathological results were confirmed by two or more pathologists,and complete clinical case data and follow-up data were available.The follow-up time was 3-60 months and the follow-up method was the combined telephone follow-up of the outpatient department,which ensured that each patient was followed up once every 3 months.Immunohistochemical technique was used to examine the expression of Oct4 protein in 89 cases of LCC tissues and 77 cases of RCC tissues.Relationship between the expression of Oct4 and clinicopathological parameters and prognostic value of Oct4 in colon cancer were analyzed.All data were statistically analyzed by Graphpad 6.0 software and SPSS20.0 software,and P < 0.05 was considered statistically significant.Compared the expression of Oct4 protein in the cancer tissue and its corresponding paracancerous tissue,the correlation between the expression level of Oct4 protein in the cancer tissue and the patient's age,tumor diameter,histological grade,lymph node metastasis,pathological classification and other clinical pathology was analyzed.Subsequently,kaplan-meier,Cox and log-rank methods were used to analyze the survival of colorectal cancer patients.Results(1)The positive rate of Oct4 protein in LCC was 68.54%,and that in RCC was71.43%,meaning no obvious differences between these two groups;In addition,Oct4 expression in RCC were positively correlated with histological grade,lymph node metastasis and Dukes staging,but in the LCC,Oct4 expression was only positively correlated with histological grade and Dukes staging.(2)In survival analysis,the 5-year survival rate of LCC was significantly higher than RCC(P<0.01);in left half colon cancer patients,no obvious correlation was found between positive and negative Oct4 expression in OS;and in right half colon cancer patients,Oct4 over-expression indicated poor prognosis(P<0.05).Also,in Cox survival analysis,Oct4 over-expression indicated poor prognosis in RCC,but not in LCC.ConclusionsThese results indicated that Oct4 might play different roles in left and right colon cancer,providing important theoretical basis for exploring new targets for the diagnosis and treatment of colon cancer.
