| Purpose:The Lewis lung cancer mouse model was used as the research object to observe the effect of moxibustion on tumor weight,relative tumor proliferation rate,tumor inhibition rate,and the expression of bcl-2 and bax protein in tumor tissues of mice.The mechanism of apoptosis provides experimental evidence.Material and method:Twenty-four male C57BL/6 mice were selected and transplanted with 0.1 mL of Lewis lung cancer cell suspension(1.0×10~7cells/mL)subcutaneously in the right armpits of mice using subcutaneous heterotopic transplantation to establish a Lewis lung cancer Transplantation tumor model.The mice were kept for 5 days and their tumorigenesis was closely observed.When the tumors grew to 50-75 mm~3,the mice were randomly divided into the model group,the cisplatin group,the moxibustion group,and the moxibustion plus cisplatin group.Group 6 only.The model group was given intraperitoneal injection of saline,0.1mL/10g,once/week for 2 weeks and administered twice;Cisplatin group was given intraperitoneally with cisplatin solution,0.05mg/10g,once/week,continued 2 Weeks,administration 2 times;moxibustion group selected the mouse"Zhongfu"point,the self-made smokeless small Moxa cone affixed to the“Zhongfu”point area,The self-produced smokeless Aifei was applied to the“Zhongfu”point area,Ignited it,and burned one,five moxa-cone/times,one time/day,continuous moxibustion treatment for 2 weeks;Cisplatin combined moxibustion group,abdominal cavity Injection of cisplatin,0.05mg/10g,once/week,two times administration,simultaneous moxibustion with Aizhi,method with moxibustion group.Continue to observe and record daily life activities,body weight changes,and tumor morphological changes.After 14 days of treatment,the mice were sacrificed and the tumor tissues were stripped to calculate the relative proliferation rate and tumor growth inhibition rate of the tumors.Western blot was used to detect the expression of bax and bcl-2 protein in each group of Lewis lung cancer tissues.Results:1.The mice in the model group had the worst survival condition,the mice were thin,the tumors were huge,and the activity was difficult.Compared with the model group,the activity of mice in the cisplatin group,moxibustion group,moxibustion plus cisplatin group was relatively good.Among the moxibustion group,the moxibustion group and the cisplatin group,the mice had the best survival condition.The tumors were smaller than the model group and the exercise was slightly delayed.The activity of the mice in the cisplatin group was compared with the moxibustion group,the moxibustion group and the cisplatin group.Compared to slightly worse.2.Mouse tumor weight changes:The tumor weights of the model group,moxibustion group,cisplatin group,and moxibustion plus cisplatin group were 19.31±2.57,11.27±1.48(g),9.18±1.18(g),and 8.86±1.81(g).The tumor weight of mice in the moxibustion group,cisplatin group,and moxibustion plus cisplatin group was lower than that in the model group,and the weight of the tumor in the moxibustion plus cisplatin group was the lightest(p<0.05).3.Tumor tissue tumor proliferation rate and tumor growth inhibition rate in lung cancer mice:Compared with the model group,the proliferation of tumors in the cisplatin group,the moxibustion group,and the moxibustion plus cisplatin group had statistical significance(P<0.01).In group comparison,the relative proliferation rates of tumors in the moxibustion group,cisplatin group,and moxibustion plus cisplatin group were 58.75%,48.35%,and46.44%,respectively,and the tumor inhibition rates were 41.64%,52.44%,and 54.13%,respectively.The treatment methods can effectively inhibit the growth of lung cancer.Among them,the moxibustion plus cisplatin group had the best inhibitory effect on tumors,followed by the cisplatin group and the moxibustion group.There was a significant difference compared with the model group(P<0.01).4.Changes in lung tissue protein expression in Lewis lung cancer model mice:Compared with the model group,bcl-2 protein expression levels in the tumor tissues of the cisplatin group,the moxibustion group,and the moxibustion plus cisplatin group significantly decreased(P<0.01).),Bax protein expression levels were significantly upregulated(P<0.01).All groups were also statistically significant,among which the moxibustion plus cisplatin group had the best effect on cell apoptosis induced by mouse tumors(P<0.01).Conclusions:1.Moxibustion“Zhongfu”acupoints can improve the quality of life of lung cancer mice.2.Moxibustion“Zhongfu”acupoints can inhibit the growth of Lewis lung tumors.3.Moxibustion“Zhongfu”acupoints can influence apoptosis-related proteins in Lewis lung cancer mice. |
