Objective:To investigate the protective effect of Tricholoma matsutake polysaccharides on PC12 cell injury induced by MPP~+.Methods:The effects of MPP~+and Tricholoma matsutake polysaccharides on cell activity were detected by MTT assay,and the morphological changes of cells in each group were observed by light microscope,and the content of oxygen free radical(ROS),malondialdehyde(MDA)and reduced glutathione(GSH)in the cells of each group was measured with ROS,MDA and GSH kits.The apoptosis of cells was detected by TUNEL and Acridine orange staining,and the mRNA and protein expression of pro-apoptosis factors Caspase-3 and Bax were detected by qRT-PCR and Western blot,respectively.Results:Compared with the normal control group,the activity of cells in the MPP~+damage model group was significantly decreased,the number of cell protrusions and apoptotic bodies were markedly reduced,the content of ROS and MDA was obviously increased,while the content of GSH was significantly decreased,and the apoptosis rate was markedly increased in the MPP~+damage model group.Compared with the MPP~+damage model group,the activity of cells in the groups of 5,50 and 500ng/mL matsutake polysaccharides was significantly increased.The number of cell protrusions and apoptotic bodies was significantly increased,and the content of ROS and MDA were decreased,while the content of GSH was obviously increased,and the apoptosis rate was decreased in thematsutake polysaccharides groups.qRT-PCR and Western blot detects showed that the expressions of Bax and Caspase-3 in the MPP~+damage model group were significantly higher than those in the normal control group.Compared with the MPP~+damage model group,the mRNA and protein expression levels of Bax and Caspase-3 in the Matsutake polysaccharide groups were markedly decreased.Conclusion:Tricholoma matsutake polysaccharides can improve MPP~+induced PC12cell injury probably though anti-oxidation and anti-apoptosis effects. |
