Objective:To study the effect of Garlic polysaccharide on cell proliferation and the protective effect of Garlic polysaccharide and selenizing product(Seleno-garlic polysaccharide) on Pheochromocytoma cells injury induced by hydrogen peroxide.Methods:1.MTT assay for cell proliferation;2.The model that H2O2 induced injury in PC12 cells was generated.Morphological changes of PC12 cells were observed under reversal phase microscope;3.Morphological changes of apoptosis were detected by Hoechst 33258 staining;4. Lactate dehydrogenase(LDH),malondialdelyde(MDA) and superoxide dismutase(SOD) in the culture medium and MDA,SOD in cells were measured by spectroscopy respectively.Results:1.Garlic polysaccharide with different concentrations could promote the cell proliferation of PC12.2.It could resist the decrease in the cell viability and the increase in the apoptotic rate induced by 25μmol/L H2O2.After exposed to 25μmol/L H2O2 for 4h,typical morphological characteristics of cells injury were observed.Both LDH and MDA levels increased significantly,while SOD level decreased significantly in H2O2 induced group compared with those in the control group.Garlic polysaccharide could decrease LDH and MDA content and increase SOD level significantly.3.Compared with Garlic polysaccharide or sodium selenite containing the same amount of Se,the seleno-garlic polysaccharide was more effective on resisting the decrease in the cell viability and increase in the apoptotic rate induced by 25μmol/L H2O2.Seleno-garlic polysaccharide was more effective on decreasing LDH and MDA content and increase SOD level significantly than Garlic polysaccharide.Conclusion:Garlic polysaccharide stimulates the proliferation of PC12.Garlic polysaccharide and Seleno-garlic polysaccharide have protective effect on PC12 cells injury induced by hydrogen peroxide in culture,which may be related to enhancing the antioxidant abilities of PC12.Compared with Garlic polysaccharide or sodium selenite,the seleno-garlic polysaccharide has more effective antioxidant abilities.
