Compatibility Analysis Of Erwuwei Songshi Pills And Study On The Preventive And Therapeutic Effects Of Reduced Prescriptions On Chronic Liver Fibrosis In Rats

Objective:At present,clinical research shows that the Tibetan medicine Ershiwuwei Songshi pills have a certain therapeutic effect on liver diseases and are widely used in Tibetan areas.However,Tibetan medicine compounds generally contain some heavy metal components.Among them,minerals such as cinnabar and turquoise have excessive content,which has a great impact on drug safety,and has been receiving attention.In this paper,by analyzing the compatibility characteristics of Tibetan medicine Ershiwuwei Songshi pills,the Ershiwuwei Songshi pills were reduced?reduced cinnabar and turquoise?,and the effects of prevention and treatment on chronic liver fibrosis in rats were compared.To explore the feasibility and effectiveness of the original prescription for the treatment of liver disease.In order to provide reference for the optimization of Tibetan medicine prescriptions and the safety of clinical medications.Methods:1.Through the examination of some Tibetan medicine classics,such as Four Medical Classics,Xin Xiu Jing Zhu Ben Cao,etc.,summarizing the flavor,nature and efficacy of the 25 herbs in Ershiwuwei Songshi pill,and analyzing the compatibility features of flavor and of nature and efficacy of the 25 herbs in Ershiwuwei Songshi pill.And I also sorted out 21 common prescriptions for the treatment of liver diseases in the Tibetan Medical formula,counting and analyzing the using frequency of the ingredient medicine of Ershiwuwei Songshi pill in the 21 Tibetan medical formula for the treatment of liver and the compatibility frequency.The compatibility characteristics of the Ershiwuwei Songshi pills and the core components of the treatment of liver diseases were discussed.2.The Ershiwuwei Songshi pills is reduced to remove the heavy metal components cinnabar and turquoise,and the original reduced cinnabar group for removing heavy metals is obtained;the original side is Ershiwuwei Songshi pills reduction of the cinnabar group;Ershiwuwei Songshi pills reduction of the Songshi group,Ershiwuwei Songshi pills reduction of the Songshi and cinnabar group.3.Fifty-six healthy male Wistar rats were randomly divided into 7 groups:Model group,Normal control group,Positive control group,Ershiwuwei Songshi pills group,Ershiwuwei Songshi pills reduction of the cinnabar group,Ershiwuwei Songshi pills reduction of the Songshi group,Ershiwuwei Songshi pills reduction of the Songshi and cinnabar group,each group contained 8 rats.Except Normal control group,other groups were subcutaneously injected with olive oil solution containing 40%CCL₄,the rat model was successfully established in the 8th week.During the course,the rats in group Model group and Normal control group were orally administered with 1.5mL normal saline,and the rest of the groups were administered through filling the stomach with accordingly dosage of drug.Rats were killed at the 8th week.Then the biochemical analysis was used to determine ALT,AST,and AL levels in serum,while enzyme-linked immuno sorbent assay?ELISA?was applied for delecting transforming growth factor-?1?TGF-?1?,hyaluronic acid?HA?,laminin?LN?,and hydroxy-proline?HYP?levels.Liver tissue morphology was observed by HE staining.Hepatic fibrosis was evaluated by Masson trichrome staining.Results:1.The results show that Ershiwuwei Songshi pills are composed of a heat clearing formula,mainly for the treatment of heat liver diseases.2.Compared with Normal control group,the levels of ALT,AST,TGF-?1,HA,LN and HYP in Model group were significantly higher;Compared with Model group,the content of AL in Ershiwuwei Songshi pills group and Ershiwuwei Songshi pills reduction of the Songshi and cinnabar group were significantly higher,the levels of TGF-?1,HA,LN and HYP in Ershiwuwei Songshi pills group and Ershiwuwei Songshi pills reduction of the Songshi and cinnabar group were significantly lower,and liver fibrosis scores were decreased.The area of hepatic fibrosis was significantly reduced.Conclusion:Our results indicated that it had certain prevention and treatment effects on rats'liver fibrosis before and after the Ershiwuwei Songshi pills reducing party.The prevention and treatment effects on rats'liver fibrosis of Ershiwuwei Songshi pills group was the best,Ershiwuwei Songshi pills reduction of the Songshi and cinnabar group was the better,Ershiwuwei Songshi pills reduction of the cinnabar group and Ershiwuwei Songshi pills reduction of the Songshi group were the worst.
