Objective To investigate the role of dihydroartemisinin(DHA)on the expression of aquaporin 9 in glioma cells and its regulating mechanism,so as to provide new ideas for new drugs targeting glioma in clinic.Methods Human glioma cell lines U87 and U251 cell lines were randomly divided into the control and DHA group.The biological behavior of glioma cells was detected by MTT assay,flow cytometry,cell scratch assay,Transwell migration and invasion assay.AQP9 expression was detected by immunofluorescence and immunoblotting.MAPK(mitogen-activated protein kinases)related proteins were detected by immunoblotting.Results The results of MTT assay indicated that DHA could inhibit the proliferation of glioma cells.Giloma cells were arrested in G2/M phase detected by flow cytometry.Cell scratch assay,Transwell migration and invasion assay results showed that DHA could inhibit migration and invasion of glioma cells;Immunofluorescence results showed that the expression of AQP9 was up-regulated in the DHA-treated group comparedwith the control group.The results of immunoblotting showed that compared with the control group,the expression of AQP9 in DHA group was up-regulated(P<0.05)and JNK-MAPK signaling pathway was activated,but p38-MAPK and ERK-MAPK signaling pathways did not change significantly.Treatment glioma cells with JNK agonist(Anisomycin),resulted in similar effects as DHA treatment,and the addition of JNK inhibitor(SP600125)partially blocked the effect of DHA on glioma cells.Conclusion DHA could inhibit the proliferation,migration and invasion of glioma cells by activating JNK-MAPK signaling pathway and upregulation of AQP9.Objective To evaluate the diagnostic accuracy of computed tomography(CT)for pelvic lymph node metastasis in muscle-invasive bladder urothelial carcinoma(MIBUC).Methods The present study retrospectively assessed the clinical and imaging characteristics of 40 MIBUC patients who underwent radical cystectomy and pelvic lymphadenectomy.The relationship between CT imaging characteristics and pathological examination for pelvic lymph node metastasis was assessed in MIBUC patients.Results 1.Compared to pathological diagnosis,CT imaging examination for pelvic lymph node metastasis,the accuracy was 90%,the sensitivity was 75%,and the specificity was 93.8%;2.The pelvic lymph node metastasis rate increased along with more advanced tumor stage(T1,0;T2,9.1%;T3,45.5%;T4,100%).Conclusion 1.CT imaging characteristics were in great importance for evaluating pelvic lymph node metastasis in MIBUC patients;2.The probability of metastatic pelvic lymph nodes significantly increased with more advanced T stages in MIBUC patients(P<0.05).
